จากการศึกษาของนักโบราณคดีพบหลักฐานการอยู่อาศัยของมนุษย์มาตั้งแต่สมัย ก่อนประวัติศาสตร์สมัยทวาราวดี และที่สำคัญที่สุด พบหลักฐานวัฒนธรรมของเขมรโบราณ กระจายอยู่ทั่วไปในจังหวัดบุรีรัมย์ ซึ่งมีทั้งปราสาทอิฐและปราสาทหินเป็นจำนวนกว่า 60 แห่ง รวมทั้งได้พบแหล่งโบราณคดีที่สำคัญ ซึ่งกำหนดอายุได้ประมาณ พุทธศตวรรษที่ 15 - 18 อยู่ทั่วไป
หลังจากสมัยของวัฒนธรรมขอมหรือเขมรโบราณแล้ว หลักฐานทางประวัติศาสตร์ของ บุรีรัมย์เริ่มมีขึ้นอีกครั้งตอนปลายสมัยกรุงศรีอยุธยา โดยปรากฏชื่อว่าเป็นเมืองขึ้นของเมือง นครราชสีมา และปรากฏชื่อต่อมาในสมัยกรุงธนบุรีถึงสมัยกรุงรัตนโกสินทร์ บุรีรัมย์มีฐานะ เป็นเมืองๆ หนึ่งจนถึง พ.ศ.2476 ได้มีการจัดระเบียบราชการบริหารส่วนภูมิภาคใหม่ จึง ได้ชื่อเป็น จังหวัดบุรีรัมย์ มาจนถึงปัจจุบันนี้
