มนตรี ตราโมท เดิมชื่อ "บุญธรรม" เกิดเมื่อ ๑๗ มิถุนายนพ.ศ. ๒๔๔๒ ที่บ้านท่าพี่เลี้ยง อำเภอเมือง จังหวัดสุพรรณบุรีเป็นบุตรนายยิ้ม นางทองอยู่และเป็นหลานชายในพระครูสัทธานุสารีมุนีวินยานุโยคสังฆวาหะ หรือหลวงพ่อลี่ ปุริสพันธ์ อดีตเจ้าอาวาสวัดสุวรรณภูมิ และเจ้าคณะจังหวัดสุพรรณบุรี ซึ่งเป็นผู้ส่งเสริมสนับสนุนการดนตรีไทยของจังหวัดสุพรรณบุรีในสมัยนั้น
ในสมัยที่มีการประกาศสำนักนายกรัฐมนตรี เรื่องการตั้งชื่อบุคคล มนตรีจึงเปลี่ยนชื่อเป็น "มนตรี" เมื่อ ๑๕ เมษายน พ.ศ. ๒๔๘๕ นามสกุล "ตราโมท" เป็นนามสกุลที่หม่อมเจ้าคำรบ ปราโมช ประทานให้ มีสำเนียงล้อนามสกุล "ปราโมช" ขององค์
