โรงเรียนสตรีนนทบุรี กำเนิดขึ้นจากความต้องการให้มีโรงเรียนสำหรับนักเรียนสตรีโดยเฉพาะในจังหวัดนนทบุรี โดยได้รับการประกาศจัดตั้งจากกระทรวงศึกษาธิการ เมื่อวันที่ 25 มีนาคม พ.ศ. 2500 เมื่อเริ่มก่อตั้งใช้ชื่อว่า "โรงเรียนสตรีประจำสุขาภิบาลนนทบุรี" ใช้อักษรย่อว่า "ส.น.บ." ตั้งอยู่ที่ศาลากลางจังหวัดนนทบุรีหลังเก่า ริมแม่น้ำเจ้าพระยา มีพิธีเปิดโรงเรียนในวันที่ 29 มีนาคม พ.ศ. 2500 มี นางลัดดาวัลย์ นนทะนาคร เป็นครูใหญ่คนแรก มีครู 3 คน ห้องเรียน 4 ห้อง นักเรียน 33 คน

สัญลักษณ์ของโรงเรียนเป็นรูปพลับพลาประดิษฐานอยู่เหนือ หมายถึง วัดพลับพลา ซึ่งที่ตั้งของโรงเรียนในปัจจุบัน ในอดีตบางส่วนเคยเป็นที่ดินของวัดพลับพลา และรูปเมฆ มีข้อความว่า "ปญญา นรานํ รตนํ ปัญญาเป็นแก้วอันประเสริฐของนรชน" ทางโรงเรียนได้กำหนดให้ดอกแก้วเป็นดอกไม้ประจำโรงเรียนสตรีนนทบุรี และกำหนดให้วันที่ 9 มกราคมของทุกปีเป็น "วันรักโรงเรียน" ตั้งแต่ พ.ศ. 2532-ปัจจุบัน

โรงเรียนสตรีนนทบุรีมีชื่อเสียงด้านการจัดการเรียนการสอนภาษาอังกฤษ ภาษาจีน ภาษาฝรั่งเศส ภาษาเยอรมัน และภาษาญี่ปุ่น และมีชื่อเสียงดีเด่นในด้านการกีฬาบาสเกตบอลและนาฏศิลป์ ได้รับคัดเลือกจากกระทรวงศึกษาธิการให้เป็นโรงเรียนดีเด่นรับรางวัลพระราชทานโรงเรียนมัธยมศึกษาประจำปี พ.ศ. 2525 พ.ศ. 2541-2549 และมีนักเรียนได้รับคัดเลือกเป็นนักเรียนรับรางวัลพระราชทานปีการศึกษา 2542-2550
