ต่อมาในปี 2457 ต้นรัชกาลที่ 6 ได้ย้ายอำเภอมาตั้งที่บ้าน “สำเพ็ง” ซึ่งเป็นย่านการค้าที่สำคัญในสมัยนั้น จนกระทั่งปี 2481 สมัยรัชกาลที่ 8 ได้เปลี่ยนชื่อจาก “อำเภอนางบวช” มาเป็น “อำเภอสามชุก” และย้ายมาตั้ง อยู่ริมลำน้ำสุพรรณบุรี (ท่าจีน) ซึ่งแยกมาจากแม่น้ำเจ้าพระยา โดยผ่านคลอง มะขามเฒ่า

อำเภอสามชุกเดิมมีพื้นที่ 774.9 ตารางกิโลเมตร ต่อมาในปี 2528 ได้มีการตั้งอำเภอหนองหญ้าไซ จึงแบ่งบางส่วนออกไป ยังคงเหลือเพียง 362 ตารางกิโลเมตร
