ไทยสถาปนาความสัมพันธ์ทางการทูตกับกัมพูชามาตั้งแต่วันที่ 19 ธันวาคม พ.ศ. 2493 ทั้งสองประเทศมีประวัติศาสตร์ วัฒนธรรม ศาสนา และรูปแบบการดำรงชีวิตของประชาชนที่คล้ายคลึงกัน มีความสัมพันธ์ ในระดับประชาชน โดยเฉพาะประชาชนบริเวณแนวชายแดนที่ใกล้ชิด และมีการแลกเปลี่ยนทางการค้าปริมาณมาก
ไทยและกัมพูชามีความร่วมมือกันในหลายด้าน เช่น มีคณะกรรมาธิการร่วมว่าด้วยความร่วมมือทวิภาคีไทย–กัมพูชา (Joint Commission on Bilateral Cooperation – JC) ซึ่งเป็นเวทีสำหรับ การเจรจาและตกลงเกี่ยวกับความร่วมมือในทุกๆ ด้าน ทั้งด้านเศรษฐกิจ
สังคม วัฒนธรรม การศึกษา การท่องเที่ยว ฯลฯ โดยได้มีการประชุม ครั้งแรกเมื่อปี พ.ศ. 2538 ณ กรุงพนมเปญ ประเทศกัมพูชา และ ครั้งล่าสุดคือการประชุม JC ครั้งที่ 7 ณ เมืองเสียมราฐ เมื่อวันที่ 3 – 4 กุมภาพันธ์ พ.ศ. 2554 ทั้งนี้ ไทยเป็นเจ้าภาพจัดการประชุม JC ครั้งที่ 8 ในปี พ.ศ. 2555
อย่างไรก็ดี จากกรอบความตกลงอาเซียนว่าด้วยการอำนวย ความสะดวกในการขนส่งข้ามแดน (ASEAN Frame Work Agreement for The Facilitation of Inter-State Transport) และความตกลง ว่าด้วยการขนส่งข้ามพรมแดนในอนุภูมิภาคลุ่มแม่น้ำโขง (Great Mekong Subregion Cross-Border Transport Agreement) ทำให้ภาครัฐ ทั้งไทยและกัมพูชาได้ประสานงานและร่วมมือกันในการลดอุปสรรคต่างๆ และลดค่าใช้จ่ายในการขนส่งสินค้าผ่านเส้นทางผ่านแดน และให้ความสำคัญ อย่างจริงจังในการทำงานร่วมกับสมาคมที่เกี่ยวข้องกับการขนส่งทางถนน แก้ไขปัญหาเรื่องรถวิ่งเที่ยวเปล่าเพื่อลดต้นทุนการขนส่งสินค้า
