โรงเรียนจุฬาภรณราชวิทยาลัย บุรีรัมย์ เป็นหนึ่งในจำนวน 12 โรงที่จัดตั้งขึ้น เนื่องในวโรกาสที่สมเด็จ พระเจ้าลูกเธอเจ้าฟ้าจุฬาภรณ วลัยลักษณ์อัครราชกุมารี ทรงพระชนมายุครบ 3 รอบ เมื่อวันที่ 4 กรกฎาคม 2536 ด้วยสำนึกในพระมหากรุณาธิคุณ ที่ทรงมีต่องาน ด้านวิทยาศาสตร์และสิ่งแวดล้อมรวมทั้งการพัฒนา คุณภาพชีวิตของประชากร ประกอบกับภาวะที่ชาติต้องการบุคลากรที่มีความรู้ ความสามารถทางด้าน วิทยาศาสตร์ เทคโนโลยีและสิ่งแวดล้อมไปช่วยพัฒนาโครงการต่างๆ ของรัฐ กรมสามัญศึกษาจึงได้จัดตั้ง โรงเรียนเพื่อปลูกฝังและส่งเสริม พร้อมทั้งให้โอกาสแก่เยาวชนที่มีความสนใจ มีความสามารถทางวิทยาศาสตร์ เทคโนโลยีและ สิ่งแวดล้อม ได้ฝึกฝนด้านนี้เป็นพิเศษตั้งแต่วัยเยาว์ โดยขอพระราชทานพระราชานุญาตขนานนามว่า “จุฬาภรณราชวิทยาลัย” และใช้นามภาษาอังกฤษว่า “Princess Chulabhorn’s College”

โรงเรียนดำเนินการรับนักเรียนครั้งแรกเมื่อวันที่ 1 มีนาคม 2537 เปิดรับนักเรียนชั้นมัธยมศึกษาชั้นปีที่ 1 และชั้นมัธยมศึกษาปีที่ 4 ชั้นละ 1 ห้องเรียน เปิดเรียนวันแรกเมื่อวันที่ 26 พฤษภาคม 2537 ซึ่งในขณะนั้นอาคารเรียน และอาคารประกอบต่างๆ อยู่ระหว่าง ดำเนินการก่อสร้างจึงใช้สถานที่เรียนชั่วคราวที่สถาบันราชภัฏบุรีรัมย์ และย้ายมาเรียนที่สถานที่ปัจจุบันเมื่อวันที่ 3 มิถุนายน 2538

โรงเรียนจุฬาภรณราชวิทยาลัย บุรีรัมย์ ได้รับบริจาคที่ดิน จำนวน 42 ไร่ 19 ตารางวา ตามโฉนดเลขที่ 24212, 31105, 31106, 31107 จากอดีต ฯพณฯ ท่าน พรเทพ เตชะไพบูลย์ เพื่อใช้เป็นสถานที่จัดตั้งโรงเรียนจุฬาภรณราชวิทยาลัย บุรีรัมย์ ประจำเขต การศึกษา 11 โดยมี นายอุทัย นิวัตินุวงศ์ รักษาการผู้ดูแลเป็นคนแรก
