พิพิธภัณฑสถานแห่งชาติ เจ้าสามพระยา เป็นพิพิธภัณฑสถานแห่งชาติ ในจังหวัดพระนครศรีอยุธยา ตั้งอยู่ที่ตำบลประตูชัย ถนนปรีดีพนมยงค์ ตรงข้ามกับมหาวิทยาลัยราชภัฏพระนครศรีอยุธยา สร้างขึ้นเนื่องในวโรกาสที่พระบาทสมเด็จพระเจ้าอยู่หัว และสมเด็จพระนางเจ้าฯ พระบรมราชินีนาถ เสด็จพระราชดำเนินมาทอดพระเนตรโบราณวัตถุที่พบจากกรุพระปรางค์ วัดราชบูรณะ จังหวัดพระนครศรีอยุธยา เมื่อวันที่ 23 พฤศจิกายน พ.ศ. 2500

พิพิธภัณฑสถานแห่งชาติ เจ้าสามพระยา นับเป็นพิพิธภัณฑสถานแห่งชาติ แห่งแรกของไทยที่มีรูปแบบการจัดแสดงแบบใหม่คือ นำโบราณวัตถุมาจัดแสดงจำนวนไม่มากจนเกินไปและใช้แสงสีมาทำให้การนำเสนอดูน่าสนใจ
ในครั้งนั้น พระบาทสมเด็จพระเจ้าอยู่หัว มีพระราชปรารภกับรัฐมนตรีกระทรวงศึกษาธิการและอธิบดีกรมศิลปากรในสมัยนั้นว่า “ โบราณวัตถุ และศิลปวัตถุที่พบในกรุพระปรางค์วัดราชบูรณะนี้ สมควรจะได้มีพิพิธภัณฑสถานเก็บรักษา และตั้งแสดงให้ประชาชนในจังหวัดพระนครศรีอยุธยานี้ หาควรนำไปเก็บรักษา และตั้งแสดง ณ ที่อื่นไม่”

ด้วยเหตุนี้ กรมศิลปากรจึงได้สร้าง พิพิธภัณฑสถานแห่งชาติ เจ้าสามพระยา ขึ้น เพื่อเก็บรักษาจัดแสดงโบราณวัตถุที่พบจากกรุพระปรางค์วัดราชบูรณะ และโบราณวัตถุพบในจังหวัดนี้ โดยใช้เงินเงินบริจาคจากประชาชน และผู้บริจาคได้รับพระพิมพ์ที่พบจากกรุพระปรางค์วัดราชบูรณะเป็นการสมนาคุณ

ทั้งนี้พระบาทสมเด็จพระเจ้าอยู่หัวทรงพระกรุณาโปรดเกล้าฯ ให้อัญเชิญพระนามสมเด็จพระบรมราชาธิราชที่ 2 (เจ้าสามพระยา) ผู้ทรงสร้างพระปรางค์วัดราชบูรณะเป็นนามพิพิธภัณฑ์ และเมื่อสร้างเสร็จแล้วพระบาทสมเด็จพระเจ้าอยู่หัว และสมเด็จพระนางเจ้าฯ พระบรมราชินีนาถ เสด็จพระราชดำเนินทรงเปิดพิพิธภัณฑสถานแห่งชาติ เมื่อวันที่ 26 ธันวาคม พ.ศ. 2504
