ต่อมาเจ้าพระยากลาโหม ซึ่งเป็นพระญาติของพระมารดาพระเจ้าทรงธรรมได้ขึ้นครองราชย์ ระหว่าง พ.ศ. ๒๑๗๓ - ๒๑๙๘ ปรากฏพระนามในพงศาวดารว่า สมเด็จพระเจ้าปราสาททอง เนื่องจากบางปะอินเป็นพระราชเคหสถานเดิมของพระองค์ จึงโปรดเกล้าฯ ให้สร้างตำหนักไอสวรรย์ทิพยอาสน์ไว้ริมสระ และสร้างวัดชุมพลนิกายารามขึ้นเมื่อ พ.ศ.๒๑๗๔ เพราะเคยเป็นที่ประชุมไพร่พลมาก่อน

ต่อมาพระบาทสมเด็จพระจอมเกล้าเจ้าอยู่หัวได้ทรงปฎิสังขรณ์วัดนี้ตามแบบเดิมทุกอย่าง และโปรดเขียนภาพชาดกไว้ที่ผนังพระอุโบสถด้วย ซึ่งยังปรากฏอยู่จนทุกวันนี้
