ปรีดี พนมยงค์ เกิดเมื่อวันที่ 11 พฤษภาคม พ.ศ. 2443 ณ เรือนแพหน้าวัดพนมยงค์ ตำบลท่าวาสุกรี อำเภอกรุงเก่า จังหวัดพระนครศรีอยุธยา ในครอบครัวชาวนาไทย เป็นบุตรคนที่ 2 จากจำนวนพี่น้อง 6 คน ของนายเสียง และนางลูกจันทน์ พนมยงค์
บรรพบุรุษของปรีดีตั้งถิ่นฐานอยู่ใกล้วัดพนมยงค์มาเป็นเวลาช้านาน โดยที่บรรพบุรุษข้างบิดานั้นสืบเชื้อสายมาจากพระนมในสมัยอาณาจักรอยุธยาชื่อ "ประยงค์" พระนมประยงค์เป็นผู้สร้างวัดในที่สวนของตัวเอง โดยตั้งชื่อวัดตามผู้สร้างว่า วัดพระนมยงค์ หรือ วัดพนมยงค์ กาลเวลาล่วงเลยมาจนเมื่อมีการประกาศพระราชบัญญัติขนานนามสกุล พ.ศ. 2456 ทายาทจึงได้ใช้นามสกุลว่า "พนมยงค์" และได้อุปถัมภ์วัดนี้เรื่อยมาจนถึงปัจจุบัน
บรรพบุรุษรุ่นปู่-ย่าของปรีดีประกอบกิจการค้าขายมีฐานะเป็นคหบดีใหญ่ แต่นายเสียงบิดาของปรีดีเป็นคนชอบชีวิตอิสระไม่ชอบประกอบอาชีพค้าขายเจริญรอยตามบรรพบุรุษ จึงหันไปยึดอาชีพกสิกรรม เริ่มต้นด้วยการทำป่าไม้ และต่อมาได้ไปบุกเบิกถางพงร้างเพื่อจับจองที่ทำนาบริเวณทุ่งหลวง อำเภอวังน้อย แต่ก็ต้องประสบกับปัญหาภัยธรรมชาติ โขลงช้างป่า และแมลงที่มารบกวนทำลายต้นข้าวทำให้ผลผลิตออกมาไม่ดี ไม่สามารถขายข้าวได้ ซ้ำร้ายรัฐบาลได้ให้สัมปทานบริษัทขุดคลองแห่งหนึ่งขุดคลองผ่านที่ดินของนายเสียงและยังเรียกเก็บค่าขุดคลอง (ดูประวัติคลองรังสิต)ซึ่งบิดาของปรีดีต้องกู้เงินมาจ่ายเป็นค่ากรอกนาในอัตราไร่ละ 4 บาท แลกกับการได้ครอบครองที่ดินที่จับจองไว้จำนวน 200 ไร่ ทำให้ฐานะทางเศรษฐกิจของครอบครัวย่ำแย่ลงไปอีก ต้องอดทนเป็นหนี้สินอยู่หลายปี เหตุการณ์ครั้งนั้นยังทำให้ราษฎรผู้บุกเบิกจับจองที่ดินมาก่อนต้องสูญเสียที่ดินไปเป็นจำนวนมากและกลายเป็นชาวนาผู้เช่าที่ในที่สุด
จากการเติบโตในครอบครัวชาวนานี้เอง ปรีดีจึงได้สัมผัสรับรู้เป็นอย่างดีถึงสภาพความเป็นอยู่และความทุกข์ยากของชนชั้นชาวนาทั้งหลายที่ฝากชีวิตไว้กับความไม่แน่นอนของดินฟ้าอากาศ ราคาพืชผลในตลาด และดอกเบี้ยของนายทุน นอกจากนี้ยังต้องพบกับการถูกเอารัดเอาเปรียบจากเจ้าที่ดินศักดินาที่กระทำผ่านการเก็บภาษีและการถือครองกรรมสิทธิ์ที่ดิน ประสบการณ์เหล่านี้เป็นแรงกระตุ้นให้ปรีดีคิดการเปลี่ยนแปลงทางสังคม เศรษฐกิจ และการเมืองของประเทศในเวลาต่อมา
