พันเอก พระยาพหลพลพยุหเสนา ชื่อเดิมว่า "พจน์ พหลโยธิน" เกิดเมื่อวันที่ ๒๙ มีนาคม ๒๔๓๐ เวลา ๐๓.๓๐ น. ณ บ้านหน้าวัดราชบูรณะ จังหวัดพระนคร เป็นบุตรของพันเอก พระยาพหลพลพยุหเสนา (ถิ่น พหลโยธิน) กับคุณหญิงจับ สมรสกับคนผู้หญิงบุญหลง พหลพลพยุหเสนา
เริ่มการศึกษาชั้นประถมศึกษาที่โรงเรียนวัดจักรวรรดิราชาวาส (วัดสามปลื้ม) และย้ายไปศึกษาต่อที่โรงเรียนสุขุมาลวิทยาลัย จนกระทั่งเข้าศึกษาต่อที่โรงเรียนนายร้อยทหารบก ต่อมาเมื่ออายุ ๑๖ ปี ได้รับทุนรัฐบาลไปศึกษาต่อที่โรงเรียนนายร้อยทหารบกในเมืองโกรสลิสเตอร์ เฟล เด ประเทศ เยอรมันนี ศึกษาอยู่ ๓ ปี ต่อจากนั้นได้เข้าประจำอยู่ในกองทัพบกเยอรมัน สังกัดกรมทหารปืนใหญ่ที่ ๔ ในปี พ.ศ.๒๔๕๕ ได้เดินทางไปศึกษาต่อวิชาช่างแสงที่ประเทศเดนมาร์ก เรียนได้ปีเดียวก็ถูกเรียกตัวกลับเนื่องจากเงินทุนการศึกษาไม่เพียงพอ
