พระบาทสมเด็จพระปรเมนทรมหาวชิราวุธ พระมงกุฎเกล้าเจ้าอยู่หัว ทรงวางศิลาฤกษ์ ตึกบัญชาการโรงเรียน ข้าราชการพลเรือนฯ (ปัจจุบันอาคารมหาจุฬาลงกรณ์) เมื่อวันที่ ๓ มกราคม ๒๔๕๘ และทรงสถาปนาโรงเรียนข้าราชการพลเรือนขึ้นเป็นจุฬาลงกรณ์มหาวิทยาลัย เมื่อวันที่ ๒๖ มีนาคม ๒๔๕๙ อีก ๑ ปีต่อมา คือในเดือนตุลาคม ๒๔๖๐ พระเจ้าน้องยาเธอกรมหมื่นชัยนาทนเรนทร ซึ่งเป็นอธิการบดีกรมมหาวิทยาลัย ในกระทรวงธรรมการพระองค์แรก ได้ทรงตั้งคณะอักษรศาสตร์และวิทยาศาสตร์ขึ้น ซึ่งเป็นหนึ่งในสี่คณะที่ได้มีการจัดตั้งขึ้น อีกสามคณะได้แก่ คณะวิศวกรรมศาสตร์ คณะแพทยศาสตร์ และคณะรัฐศาสตร์

ต่อมาเมื่อ พ.ศ. ๒๔๗๖ ได้มีพระราชกฤษฎีกา แยกคณะอักษรศาสตร์และวิทยาศาสตร์ออกเป็นสองคณะต่างหากจากกัน คณะอักษรศาสตร์แบ่งออกเป็นแผนกอักษรศาสตร์ และแผนกฝึกหัดครู คณะวิทยาศาสตร์แบ่งออกเป็นแผนกวิทยาศาสตร์ และแผนกฝึกหัดครู แต่ในปีเดียวกันนั้นเอง คณะอักษรศาสตร์และคณะวิทยาศาสตร์ ได้กลับยุบรวมกันเป็นคณะเดียวอีก โดยแบ่งงานออกเป็น ๙ แผนกคือ แผนกสารบรรณและหอสมุดของคณะ แผนกเคมี แผนกฟิสิกส์ แผนกชีววิทยา แผนกคณิตศาสตร์ แผนกภาษาไทยและโบราณตะวันออก แผนกภาษาปัจจุบัน แผนกภูมิศาสตร์และประวัติศาสตร์ แผนกฝึกครู
ในด้านการสอนมีการปรับปรุงหลักสูตรให้สอนถึงขั้นปริญญาบัณฑิตเมื่อ พ.ศ. ๒๔๗๗ อักษรศาสตรบัณฑิตรุ่นแรกที่สำเร็จการศึกษาเมื่อ พ.ศ. ๒๔๗๘ มีจำนวน ๓๓ คน ต่อมาในปี พ.ศ. ๒๔๘๕ ได้เปิดสอนในขั้นปริญญามหาบัณฑิต มีผู้สำเร็จการศึกษาได้รับปริญญาอักษรศาสตรมหาบัณฑิตเป็นรุ่นแรกใน พ.ศ. ๒๔๘๗ จำนวน ๒ คน
