เมื่อทรงพระเยาว์ ทรงบรรชาเป็นสามเณร ณ วัดพระศรีรัตนศาสดาราม เมื่อวันที่ 20 สิงหาคม พ.ศ. 2447 ทรงศึกษา ณ โรงเรียนราชกุมาร ในพระบรมมหาราชวัง และทรงเสด็จไปศึกษาต่อต่างประเทศที่ "โรงเรียน แฮร์โรว์" ในประเทศอังกฤษ และวิชาการทหารเรือในประเทศเยอรมัน เมื่อจบการศึกษาได้รับพระราชอิสริยยศเป็น นายเรือตรีแห่งราชนาวีเยอรมัน และนายเรือตรีแห่งราชนาวีสยาม แล้วเสด็จกลับมาทรงรับราชการในราชนาวีสยามอยู่ระยะหนึ่ง ทรงลาออกจากกองทัพเรือเมื่อ พ.ศ. 2459 แล้วเสด็จไปศึกษาวิชาการสาธารณสุขและวิชาทางการแพทย์ ณ มหาวิทยาลัยฮาร์วาร์ด สหรัฐอเมริกา ทรงสอบได้ประกาศนียบัตรการสาธารณสุข และ ปริญญาแพทยศาสตรบัณฑิตชั้น Cum Laude และทรงได้รับการแต่งตั้งเป็นสมาชิกสมาคมเกียรตินิยมทางการศึกษาแพทย์ อัลฟา โอเมกา อัลฟา ด้วย
ตำแหน่งราชการ ทรงเป็นอธิบดีกรมมหาวิทยาลัย กระทรวงศึกษาธิการ ข้าหลวงตรวจการศึกษาทั่วไป นายกกรรมการคณะแพทยศาสตร์และศิริราชพยาบาล กรรมการสภากาชาดสยาม ประธานกรรมการอำนวยการวชิรพยาบาล พระอาจารย์พิเศษคณะอักษรศาสตร์และวิทยาศาสตร์ และแพทย์ประจำโรงพยาบาลแมคคอร์มิค
ทรงอภิเษกสมรสกับ นางสาวสังวาลย์ตะละภัฏ (สมเด็จพระศรีนครินทรา บรมราชชนนี) เมื่อวันที่ 10 กันยายน พ.ศ.2463 ณ วังสระปทุม มีพระราชโอรส และพระธิดา รวม 3 พระองค์ ได้แก่
1. สมเด็จพระเจ้าพี่นางเธอ เจ้าฟ้ากัลยาณิวัฒนา กรมหลวงนราธิวาสราชนครินทร์ ประสูติ ณ กรุงลอนดอน ประเทศอังกฤษ เมื่อวันที่ 6 พฤษภาคม พ.ศ.2466
