ประยุทธ์ จันทร์โอชา เกิดเมื่อวันที่ 21 มีนาคม พ.ศ. 2497 ที่จังหวัดนครราชสีมา มีชื่อเล่นว่า ตู่ หรือที่สื่อมวลชนนิยมเรียกว่า "บิ๊กตู่" จบการศึกษาระดับมัธยมจากโรงเรียนวัดนวลนรดิศ กรุงเทพมหานคร จบโรงเรียนเตรียมทหาร รุ่นที่ 12 และโรงเรียนนายร้อยพระจุลจอมเกล้า รุ่นที่ 23
สมรสกับรองศาสตราจารย์ นราพร จันทร์โอชา อดีตอาจารย์ประจำสถาบันภาษา จุฬาลงกรณ์มหาวิทยาลัย
นอกจากนั้นพล.อ.ประยุทธ์ เป็นพี่ชายของพลโท ปรีชา จันทร์โอชา แม่ทัพภาคที่ 3
การทำงาน
ชีวิตราชการทหารรับราชการอยู่ที่กรมทหารราบที่ 21 รักษาพระองค์ หรือ "ทหารเสือราชีนี" มาโดยตลอด โดยเริ่มมาจากตำแหน่งผู้บังคับการกองพัน จนถึงผู้บังคับการกรม จากนั้นจึงย้ายไปอยู่ที่กองพลทหารราบที่ 2 รักษาพระองค์ และรับตำแหน่งรองแม่ทัพภาคที่ 1
ในเหตุการณ์รัฐประหารโดยคณะปฏิรูปการปกครองในระบอบประชาธิปไตยอันมีพระมหากษัตริย์ทรงเป็นประมุข (คปค.) เมื่อวันที่ 19 กันยายน พ.ศ. 2549 ที่มีพล.อ.สนธิ บุญยรัตกลิน เป็นหัวหน้าคณะ พล.อ.ประยุทธ์ จันทร์โอชา ในขณะนั้นมียศเป็น "พลตรี" ก็เป็นผู้หนึ่งที่มีบทบาทสำคัญในการยึดอำนาจด้วยรับคำสั่งตรงจาก พลโทอนุพงษ์ เผ่าจินดา แม่ทัพภาคที่ 1 หลังจากนั้นเมื่อ พล.ท.อนุพงษ์ได้รับแต่งตั้งให้เป็นผู้ช่วยผู้บัญชาการทหารบกและเลื่อนชั้นยศเป็น "พลเอก" พลตรีประยุทธ์ก็ได้เลื่อนชั้นยศขึ้นเป็น "พลโท" และรับตำแหน่งแม่ทัพภาคที่ 1 และได้รับแต่งตั้งให้เป็นสมาชิกสภานิติบัญญัติแห่งชาติจากคณะมนตรีความมั่นคงแห่งชาติ (คมช.) อีกด้วย
พล.อ.ประยุทธ์ เป็นนายทหารที่มีความสนิทสนมกับพล.อ.อนุพงษ์ เป็นอย่างมาก ด้วยความเป็นผู้ใต้บังคับบัญชามาตลอดในกรมทหารราบที่ 21 รักษาพระองค์ โดยพลเอกประยุทธ์นับถือพลเอกอนุพงษ์เสมือนพี่และอาจารย์คนหนึ่งของตน โดยพลเอกประยุทธ์เป็นนายทหารที่มีบุคลิกที่อ่อนนุ่ม โดยมักติดคำว่า "นะจ๊ะ" ต่อท้ายการพูด จึงได้รับอีกชื่อหนึ่งจากสื่อมวลชนว่า "ตู่นะจ๊ะ"
พล.อ.ประยุทธ์ ได้รับตำแหน่งเป็น รองหัวหน้าผู้รับผิดชอบในการแก้ไขสถานการณ์ฉุกเฉิน ระหว่างวันที่ 2 กันยายน พ.ศ. 2551 ถึง 14 กันยายน พ.ศ. 2551 และรับตำแหน่งรองผู้อำนวยการกองอำนวยการแก้ไขสถานการณ์ฉุกเฉิน

นายอภิสิทธิ์ เวชชาชีวะ นายกรัฐมนตรีได้แต่งตั้งให้ พล.อ.ประยุทธ์ จันทร์โอชา เป็น หัวหน้าผู้รับผิดชอบในการแก้ไขสถานการณ์ฉุกเฉินตั้งแต่วันที่ 1 ตุลาคม พ.ศ. 2553 ถึง 22 ธันวาคม พ.ศ. 2554 และเป็นหนึ่งในคณะดำเนินคดีศาลยุติธรรมระหว่างประเทศระหว่างประเทศไทยและกัมพูชา พ.ศ. 2554
