เดิมศาสตราจารย์ หม่อมเจ้าสุภัทรดิศ ดิศกุล ทรงวางอนาคตไปทางสายอาชีพครู จึงทรงศึกษาวิชาครูไว้ก่อนเข้ารับราชการ แต่เมื่อทรงรับราชการในกองโบราณคดี กรมศิลปากร ก็ทรงมีภารกิจเกี่ยวข้องกับงานด้านศิลปะ ประวัติศาสตร์ และโบราณคดี ที่โปรดปราน ทั้งยั้งมีโอกาสได้ทรงศึกษาวิชาเฉพาะทางคือ ประวัติศาสตร์ศิลป์ และโบราณคดีในภาษาต่างประเทศ ดังนั้น เมื่อรวมความรู้จากการศึกษา ประสบการณ์ในการทำงาน และการที่ทรงค้นคว้าเพิ่มเติมส่วนพระองค์ตลอดเวลา จึงทำให้เป็นเลิศในวิชาการด้านประวัติศาสตร์ศิลป์ นับว่าเป็นนักปราชญ์พระองค์หนึ่ง ได้ทรงนำความรู้ทั้งหมดมาสร้างประโยชน์แก่วงการศึกษาไทย ดังนี้

พ.ศ. 2499 ศาสตราจารย์ หม่อมเจ้าสุภัทรดิศ ดิศกุล ได้ก่อตั้งการสอนประวัติศาสตร์ศิลปะ และโบราณคดี เป็นครั้งแรกในโรงเรียนศิลปศึกษาของกรมศิลปากร ทรงวางหลักสูตรการเรียนวิชานี้เช่นเดียวกับโรงเรียนลูฟร์ นักศึกษาต้องเรียนประวัติศาสตร์ทั่วไปของประเทศไทย และประเทศใกล้เคียง เช่น อินเดีย ศรีลังกา อินโดนีเซีย กัมพูชา จีน และญี่ปุ่น ในการนี้ต้องทรงจัดทำตำราเรียนให้ด้วย เพราะยังไม่มีตำราภาษาไทยด้านนี้โดยตรง ทรงอุตสาหะแปลตำราจากภาษาต่างประเทศซึ่งส่วนใหญ่เป็นภาษาฝรั่งเศส ทั้งยังทรงรับเป็นผู้สอนนักศึกษาด้วย

พ.ศ. 2507 ศาสตราจารย์ หม่อมเจ้าสุภัทรดิศ ดิศกุล ทรงย้ายจากกรมศิลปากรไปดำรงตำแหน่งอธิการบดีบัณฑิตวิทยาลัย มหาวิทยาลัยศิลปากร ตั้งแต่ พ.ศ. 2519-2524 และดำรงตำแหน่งอธิการบดี มหาวิทยาลัยศิลปากร ตั้งแต่ พ.ศ. 2525-2529 หลังจากทรงเกษียณอายุจากมหาวิทยาลัยศิลปากรแล้ว ศาสตราจารย์ หม่อมเจ้าสุภัทรดิศ ดิศกุล ทรงได้รับแต่งตั้งให้ดำรงตำแหน่งผู้อำนวยการศูนย์ภูมิภาคโบราณคดีและวิจิตรศิลป์ของเอเชียตะวันออกเฉียงใต้ (SPAFA) ในองค์การรัฐมนตรีศึกษาแห่งเอเชียตะวันออกเฉียงใต้ (SEAMEO) ตั้ง พ.ศ. 2530-2535 เมื่อพ้นจากตำแหน่งนี้แล้วทรงได้รับเชิญเป็นที่ปรึกษาด้านโบราณคดีและพิพิธภัณฑสถานของกรมศิลปากร และยังทรงรับเป็นอาจารย์พิเศษแก่สถาบันต่างๆ ทรงยุติงานค้นคว้าและงานสอนที่ทรงรักทั้งหมดลงใน พ.ศ. 2540 เนื่องจากทรงประสบอุบัติเหตุทำให้ประชวรหนัก

ศาสตราจารย์ หม่อมเจ้าสุภัทรดิศ ดิศกุลทรงได้รับการขนานพระนามจากบรรดาสานุศิษย์ในมหาวิทยาลัยศิลปากรว่า ท่านอาจารย์ เป็นพระนามที่แสดงถึงความเคารพยกย่องและสนิทสนมรักใคร่ เพราะทรงเมตตาศิษย์อย่างเสมอภาคและวางพระองค์เรียบง่ายไม่ถือยศศักดิ์ สามารถเข้ากับบุคคลทั่วไปได้ทุกระดับชั้น ทรงเต็มพระทัยที่จะถ่ายทอดความรู้และเป็นที่ปรึกษาทางวิชาการ ทั้งยังประทานคำแนะนำและแหล่งข้อมูลต่างๆ เพื่อการศึกษาเพิ่มเติมได้อย่างถูกต้องแม่นยำ ซึ่งมาจากการที่ได้ทรงศึกษาค้นคว้าจากหนังสือต่างๆ อยู่ตลอดเวลา ทรงนิพนธ์ตำราและบทความวิชาการด้านประวัติศาสตร์ ประวัติศาสตร์ศิลปะ และโบราณคดีเป็นจำนวนมาก เช่น ศิลปะอินเดีย ศิลปะชวา ศิลปะขอม ศิลปะในประเทศไทย เทวรูปสัมฤทธิ์สมัยสุโขทัย เที่ยวเมืองลังกา ศิลปะอินโดนีเซียโบราณ ประติมากรรมขอม ศาสนาพราหมณ์ในอาณาจักรขอม หนังสือนำชมพิพิธภัณฑสถานหลายแห่ง Art in Thailand, A Brief History และ Thailand ในชุด Archeaological Mundi ซึ่งตีพิมพ์ในประเทศสวิตเซอร์แลนด์เป็นภาษาอังกฤษ ภาษาฝรั่งเศสและภาษาเยอรมัน เป็นต้น นอกจากทรงนิพนธ์ตำราความรู้ทางวิชาการแล้ว ยังทรงจัดหาหนังสือทางวิชาการต่าง ๆ มาประทานแก่ห้องสมุดของคณะโบราณคดีตลอดจนห้องสมุดขององค์การสปาฟา (SPAFA) เพื่อให้เกิดประโยชน์แก่การศึกษาด้านประวัติศาสตร์ ประวัติศาสตร์ศิลปะ และโบราณคดียิ่งขึ้นด้วย
