นายเปลื้อง ฉายรัศมี เป็นบุตรของ นายคง นางนาง ฉายรัศมี

เกิดเมื่อวันที่ 25 ตุลาคม พ.ศ. 2475 ที่บ้านเลขที่ 7 บ้านนา ตำบลม่วงนา อำเภอเมืองกาฬสินธุ์ จังหวัดกาฬสินธุ์
การศึกษา จบชั้นประถมศึกษาปีที่ 4 จากโรงเรียนบ้านนา ถิ่นกำเนิด
ปัจจุบันพักอยู่บ้านเลขที่ 157 หมู่ 13 ตำบลเหนือ บ้านโพนทอง อำเภอเมืองกาฬสินธุ์ จังหวัดกาฬสินธุ์ และทำงานอยู่ที่วิทยาลัยนาฏศิลป์กาฬสินธุ์
นายเปลื้อง ฉายรัศมี แต่งงานครั้งสุดท้าย พ.ศ 2519 กับนางยุพิน ฉายรัศมี ซึ่งมีอาชีพทำนา มีบุตรชาย 2 คน และหญิง 1 คน
เมื่ออายุ 27 ปี นายเปลื้อง ฉายรัศมี ได้เข้าทำงานเป็นลูกจ้างชั่วคราว อยู่ที่โครงการเขื่อนลำปาว (พ.ศ. 2502 – 2510)
ต่อมา พ.ศ. 2516 ได้สอบบรรจุเป็นลูกจ้างประจำ อยู่ในสำนักงานป่าไม้จังหวัดกาฬสินธุ์ จนถึง พ.ศ. 2519 จึงได้ลาออก
ต่อมาปี พ.ศ. 2520 ได้เข้าทำงานที่ชลประทานจังหวัดอุบลราชธานี
ต่อมาปี พ.ศ. 2521 ได้ย้ายไปอยู่ที่จังหวัดนครพนม ถึงปี พ.ศ. 2524 ได้ย้ายมาทำงานที่อ่างเก็บน้ำห้วยมโน บ้านนา อำเภอเขาวง จังหวัดกาฬสินธุ์

ในด้านผลงานนั้น นายเปลื้อง ฉายรัศมี เป็นผู้ที่มีความสามารถพิเศษทางด้านดนตรีพื้นบ้านอีสาน สามารถเล่นและสอนถ่ายทอดได้ทั้ง พิณ แคน ซอ โปงลาง และเครื่องดนตรีอื่นแทบทุกชนิดในภาคอีสาน โดยเฉพาะโปงลางนั้น มีความสามารถเล่นได้ดีเป็นพิเศษ ทั้งนี้เพราะเป็นผู้ที่วิวัฒนาการดนตรีชนิดนี้ขึ้นมา

จนทำให้ครูเปลื้อง ฉายรัศมี ได้รับรางวัลศิลปินแห่งชาติ สาขาศิลปะการแสดง (นาฏศิลป์ไทย) สาขาย่อย ดนตรีพื้นบ้าน ปี 2529
