สำนักหอสมุด มหาวิทยาลัยกรุงเทพ เดิมมีชื่อเรียกว่า ห้องสมุดวิทยาลัยไทยเทคนิค เริ่มก่อตั้งขึ้นพร้อมกับการก่อตั้งวิทยาลัยไทยเทคนิคในปี พ.ศ. 2505 ต่อมาเมื่อมีการเปลี่ยนชื่อเรียกสถาบันจากวิทยาลัยไทยเทคนิค เป็นวิทยาลัยกรุงเทพ ห้องสมุดจึงได้เปลี่ยนชื่อเป็น แผนกห้องสมุดวิทยาลัยกรุงเทพ

ระยะแรกของการเปิดดำเนินการนั้น ห้องสมุดตั้งอยู่ ณ อาคาร 1 ชั้น 1 วิทยาเขตกล้วยน้ำไท มีเนื้อที่ประมาณ 3 ห้องเรียน เมื่อได้มีการขยายพื้นที่ตามจำนวนนักศึกษาและคณาจารย์ที่เพิ่มมากขึ้นอย่างต่อเนื่อง ในปี พ.ศ. 2509 ห้องสมุดได้ย้ายที่ทำการมา ณ อาคาร 2 (อาคาร สวัสดิ์-ล้อม) และขยายเพิ่มอีกเป็นขนาด 4 ห้องเรียน
อาคารหอสมุดสุรัตน์ โอสถานุเคราะห์ มีพื้นที่ทั้งสิ้น 23,600 ตารางเมตร เป็นอาคารประหยัดพลังงานและรักษาสิ่งแวดล้อม มีการนำอุปกรณ์เทคโนโลยีที่ทันสมัยมาใช้ในการควบคุมบริหารความปลอดภัย และได้รับการออกแบบโดยคำนึงถึงผู้พิการ เพื่อให้สามารถใช้บริการได้สะดวก ในส่วนของสำนักหอสมุดมีพื้นที่ 15,508 ตารางเมตร นอกจากนี้ยังเป็นที่ตั้งของ Cyber Center ที่พร้อมด้วยเทคโนโลยีและอุปกรณ์ที่ทันสมัย เพื่อให้บริการทางการศึกษาและรองรับความต้องการของคณาจารย์ นักศึกษา และเพื่อส่งเสริมการพัฒนาการเรียนการสอนผ่านระบบเครือข่าย (Education on Demand)

อาคารหอสมุดสุรัตน์ โอสถานุเคราะห์ เป็นที่ตั้งของ หอเกียรติคุณ (Hall of Recognition) โดยตั้งอยู่ชั้น 2 ซึ่งจัดตั้งขึ้นเพื่อรำลึกถึงบุคคลสำคัญและผู้มีพระคุณของมหาวิทยาลัยกรุงเทพ ได้แก่ ผู้อุปถัมภ์ ผู้ก่อตั้ง อดีตนายกสภามหาวิทยาลัย นายกสภามหาวิทยาลัย อธิการบดีกิตติคุณก่อตั้ง อธิการบดีกิตติคุณ อธิการบดี และผู้มีอุปการคุณ อีกทั้งยังเป็นสถานที่รวบรวมของที่ระลึก ภาพถ่าย ภาพวาด รูปปั้นของบุคคลสำคัญ และบริเวณชั้น 3 เป็นที่ตั้งของหอประวัติมหาวิทยาลัยกรุงเทพ ซึ่งจัดตั้งขึ้นเนื่องในโอกาสครบรอบ 40 ปี ในปี พ.ศ. 2545 เพื่อเป็นแหล่งรวบรวมและอนุรักษ์ทรัพยากร ของที่ระลึก สื่อความรู้และเอกสารต่าง ๆ ที่สามารถให้ข้อมูลในเรื่องที่มีคุณค่าเกี่ยวกับประวัติศาสตร์และพัฒนาการของมหาวิทยาลัยกรุงเทพ

ในวันที่ 29 พฤศจิกายน พ.ศ. 2545 สมเด็จพระเทพรัตนราชสุดาฯ สยามบรมราชกุมารี เสด็จพระราชดำเนินทรงเปิดอาคารหอสมุดสุรัตน์ โอสถานุเคราะห์ อย่างเป็นทางการ ซึ่งถือเป็นสิริมงคล อย่างยิ่ง และด้วยสถาปัตยกรรมและเทคโนโลยีที่ทันสมัย สำนักหอสมุดจึงได้รับความสนใจเข้าชมและศึกษาดูงานทั้งหน่วยงานจากภาครัฐและเอกชนอย่างต่อเนื่อง
ปี พ.ศ. 2547 อาคารหอสมุดสุรัตน์ โอสถานุเคราะห์ ได้รับรางวัลสถาปัตยกรรมดีเด่น จากสมาคมสถาปนิกสยาม ในพระบรมราชูปถัมภ์ ซึ่งถือเป็นเกียรติอย่างยิ่งของมหาวิทยาลัย
