รัชสมัยพระบาทสมเด็จพระจุลจอมเกล้าเจ้าอยู่หัว ในสมัยนั้นทางราชการเห็นว่าป้อมมีความสำคัญน้อยมาก ไม่เหมาะสมกับความก้าวหน้า ทางอาวุธยุทโธปกรณ์สมัยใหม่ จึงได้รื้อป้อมพิฆาตข้าศึก และกำแพงลง แล้วตั้งกองโรงเรียนพลทหารเรือที่ 1 ขึ้นแทน โดยได้โปรดเกล้า ฯ ให้นายพลเรือเอกพระยามหาโยธา (ฉ่าง แสงชูโต) มาอำนวยการสร้างโรงเรียนดังกล่าว สำหรับฝึกอบรมทหารใหม่ที่เกณฑ์จากชายฉกรรจ์ในจังหวัดสมุทรสงครามจำนวนสองกองร้อย ฝึกหัดหนึ่งปีแล้วส่งผลัดเปลี่ยนทหารเก่าในกรุงเทพ ฯ ตั้งมาจนถึงปี พ.ศ.2465 ในรัชสมัยพระบาทสมเด็จพระมงกุฎเกล้าเจ้าอยู่หัว ได้ยุบกองโรงเรียนพลทหารเรือ โดยกระทรวงทหารเรือได้ยกอาคาร และที่ดินให้กระทรวงมหาดไทย ใช้เป็นที่ตั้งศาลากลางจังหวัดสมุทรสงคราม ตั้งแต่ปลายปี พ.ศ.2468 จนถึงปี พ.ศ.2499 จึงได้ทำการปลูกสร้างศาลากลางจังหวัดใหม่บริเวณที่อยู่ปัจจุบัน แล้วโอนศาลากลางเดิมให้กระทรวงสาธารณสุขใช้ในกิจการโรงพยาบาลจังหวัด
ในปี พ.ศ.2437 ได้โปรดเกล้า ฯ ให้รวมหัวเมืองราชบุรี กาญจนบุรี เพชรบุรี ปราณบุรี ประจวบ ฯ และเมืองสมุทรสงครามเข้าเป็นมณฑลราชบุรี
ในปี พ.ศ.2458 ได้ยกแขวงบางช้างขึ้นเป็นอำเภอชื่ออำเภออัมพวา ตามชื่อของบ้านอัมพวา ส่วนบ้านบางช้างเป็นตำบลหนึ่งในเขตปกครองของอำเภออัมพวา
