เป็นวัดโบราณ สร้างในสมัยอยุธยา เดิมเรียกว่าวัดสลัก ตั้งอยู่กึ่งกลางระหว่างพระบรมมหาราชวังกับพระราชวังบวรสถานมงคล เป็นที่ตั้งของ “จุฬาลงกรณ์ราชวิทยาลัย” ซึ่งเป็นสถานศึกษาชั้นอุดมศึกษาของพระสงฆ์ สมเด็จพระบวรราชเจ้ามหาสุรสิงหนาทโปรดให้บูรณปฏิสังขรณ์วัดสลักเมื่อ พ.ศ. ๒๓๒๖ พร้อมกับการก่อสร้างพระราชวังบวรสถานมงคล จากนั้นทรงเปลี่ยนชื่อวัดจากวัดสลัก เป็น วัดนิพพานารามเมื่อพระบาทสมเด็จพระพุทธยอดฟ้าจุฬาโลกมหาราชโปรดเกล้าฯให้ใช้วัดนิพพานารามเป็นสถานที่สังคายนาพระไตรปิฎกใน พ.ศ. ๒๓๓๑ ได้ทรงพระกรุณาโปรดเกล้าฯ พระราชทานนามวัดใหม่ว่า “วัดพระศรีสรรเพชญ” และใน พ.ศ. ๒๓๔๖ พระราชทานนามใหม่ว่า วัดพระศรีรัตนมหาธาตุราชวรมหาวิหาร เป็นที่ประดิษฐานพระบรมสารีริกธาตุและเป็นที่ประทับของสมเด็จพระสังฆราช ใน พ.ศ. ๒๔๓๙ โปรดเกล้าฯ พระราชทานพระราชทรัพย์ส่วนพระองค์ของสมเด็จพระบรมโอรสาธิราชฯ สยามมกุฎราชกุมาร ในการบูรณะวัดมหาธาตุและพระราชทานนามว่า “วัดมหาธาตุยุวราชรังสฤษฎิ์” เป็นพระอารามหลวงชั้นเอก สังกัดคณะสงฆ์ มหานิกาย
