พิพิธภัณฑสถานแห่งชาติร้อยเอ็ด ตั้งอยู่บนถนนเพลินจิต ตำลในเมือง อำเภอเมือง จังหวัดร้อยเอ็ด จัดตั้งขึ้นตามโครงการการจัดตั้งพิพิธภัณฑสถานแห่งชาติประจำเมืองเป็นสถานที่จัดแสดงและรวบรวมเรื่องราวน่ารู้ทุกด้านของจังหวัดร้อยเอ็ด

เดิมพิพิธภัณฑสถานแห่งชาติร้อยเอ็ดจัดตั้งขึ้นตามดำริของท่านศาสตราจารย์ ดร.ก่อ สวัสดิ์พาณิชย์

โดยมีความตั้งใจที่จะให้เป็นพิพิธภัณฑ์ฯ ศิลปหัตถกรรมอีสาน โดยเฉพาะผ้าไหม และ ผ้าพื้นเมือง ต่อมาเมื่อกรมศิลปกรมีนโยบายในการจัดตั้งพิพิธภัณฑสถานแห่งชาติประจำเมืองจึงได้ทำการปรับปรุงเพิ่มเติมเนื้อหาในการจัดแสดงให้ครอบคลุมข้อมูลเรื่องราวของจังหวัดทุกด้านทั้งด้านภูมิศาสตร์ทรัพยากรธรณี โบราณคดี ประวัติศาสตร์ บุคคลสำคัญ วิถีชีวิต ประเพณี และศิลปหัตถรรม

พิพิธภัณฑสถานแห่งชาติร้อยเอ็ด เริ่มจัดตั้งมาตั้งแต่งบประมาณปี พ.ศ. 2536 และก่อสร้างอาคารต่อเนื่อง มาจนถึงปี พ.ศ.2540 จึงดำเนินการได้แล้วเสร็จสมบูรณ์ประกอบด้วยงบประมาณด้านการจัด แสดงนิทรรศการถาวร ปรับสภาพภูมิทัศน์ติดตั้งระบบไฟฟ้าและระบบปรับอากาศ จัดทำห้องประชุมและนิทรรศการพิเศษ รวมถึง ปรับปรุงบ้านพักเจ้าหน้าที่

ทั้งนี้โดยเฉพาะในส่วนของการจัดแสดงได้มีการนำเอาเทคโนโลยีสมัยใหม่มาใช้ คือ ระบบสารสนเทศ การทำหุ่นจำลองและฉากชีวิตต่าง ๆ เข้ามาประกอบการนำเสนอเรื่องราว ทำให้อาจกล่าวได้ว่าเป็นพิพิธภัณฑสถานแห่งชาติที่ทันสมัยที่สุดแห่งหนึ่งในความดูแลของกรมศิลปากร

นักท่องเที่ยวที่สนใจมาเยือน สามารถเข้าชมได้เวลา 09.00 – 16.00 น. ในวันพุธถึงวันอาทิตย์ โดยจะ ปิดทำการวันจันทร์ วันอังคารและวันหยุดนักขัตฤกษ์ และค่าธรรมเนียมในการเข้าชมสำหรับ ชาวไทย 10 บาท ชาวต่างประเทศ 30 บาท นักเรียน นักศึกษาในเครื่องแบบ ภิกษุ สามเณร และนักบวชในศาสนาต่างๆ ไม่เสียค่าธรรมเนียมเข้าชมแต่อย่างใด
