ตั้งอยู่ริมถนนโพศรี ใกล้วัดโพธิสมภรณ์ ตัวอาคารสร้างขึ้นเมื่อ พ.ศ.2463 โดยดำริพระยาศรีสุริยราชวรานุวัตร สมุหเทศาภิบาล สำเร็จราชการมณฑลอุดร คุณหญิง ข้าราชการ พ่อค้า และประชาชนในจังหวัด เพื่อใช้เป็นอาคารเรียนสำหรับ โรงเรียนนารีอุปถัมภ์ สร้างเสร็จเมื่อ พ.ศ.2468 ต่อมาพระบาทสมเด็จพระมงกุฏเกล้าเจ้าอยู่หัว โปรดเกล้าฯพระราชทานนามโรงเรียนขึ้นใหม่ ว่า "ราชินูทิศ" จึงเป็นชื่อเรียกอาคารหลังนี้สืบมา
อาคารราชินูทิศ ได้ใช้เป็นอาคารสำนักงานส่งเสริมวัฒนธรรมหญิง เมื่อ พ.ศ.2473 และเมื่อปี พ.ศ.2503 ใช้เป็นสำนักงานโครงการพัฒนาการศึกษาส่วนภูมิภาค จนต่อมา พ.ศ.2516 ได้ใช้เป็นอาคารสำนักงานศึกษาธิการเขต เขตการศึกษา 9 และสำนักงานวัฒนธรรมจังหวัดอุดรธานี ตามลำดับ อาคารราชินูทิศเป็นอาคาร 2 ชั้น ก่อด้วยอิฐหรือปูน รูปทรงแบบตะวันตก หลังคาทรงปั้นหยา มีมุขยื่นออกมาด้านหน้า ซุ้มประตูหน้าต่างโค้ง
ปัจจุบันได้ปรับปรุงอาคารดังกล่าวนี้เป็นพิพิธภัณฑ์ เมืองอุดรธานี จัดแสดงเรื่องราวต่าง ๆ เกี่ยวกับจังหวัวดอุดรธานี นับตั้งแต่ประวัติศาสตร์ โบราณคดี> ธรรมชาติวิทยา ธรณีวิทยา ประวัติศาสตร์ท้องถิ่น และศิลปวัฒนธรรม รวมถึงพระประวัติและ พระเกียรติคุณของกรมหลวงประจักษ์ศิลปาคม ผู้ก่อตั้งเมืองอุดรธานี...
