เปิดทำการสอนเมื่อวันที่ ๒๔ มิถุนายน พ.ศ. ๒๔๘๕ โดยมี พันเอกหลวงอาจศรศิลป์ (นายประพันธ์ ธนพุทธิ) ข้าหลวงตรวจการมหาดไทยภาค ๓ เป็นประธานในพิธี นายธวัชชัย พุกประยูร เป็นศึกษาธิการจังหวัด คุณหญิงเอื้อนทิพย์ เปรมโยธิน เป็นครูใหญ่คนแรก ในปีการศึกษานี้เปิดรับนักเรียนชั้นอนุบาลปีที่ ๑ จำนวน ๓๐ คน ได้เช่าอาคารเอกชนเปิดทำการสอนที่ถนนอัษฎางค์ ตำบลในเมือง อำเภอเมือง จังหวัดนครราชสีมา

คุณหญิงเอื้อนทิพย์ เปรมโยธิน ลาออกจากราชการ ศึกษาธิการจังหวัดจึงแต่งตั้งให้ นางสมศรี ศรจิตติ เป็นครูใหญ่โรงเรียนอนุบาลนครราชสีมาอีกตำแหน่งหนึ่ง

พ.ศ. ๒๔๙๐ นางสมศรี ศรจิตติ ลาออกจากราชการ เจ้าของอาคารเลิกสัญญาเช่า ศึกษาธิการจังหวัดไปเช่าบ้านเอกชน ที่ถนนจอมพล เป็นสถานที่ของโรงเรียนการเรือน และโรงเรียนอนุบาลนครราชสีมาแต่งตั้ง นางวัลภา นุกูลชิต เป็นครูใหญ่ทั้ง ๒ โรงเรียน

พ.ศ. ๒๔๙๖ ได้รับงบประมาณสร้างอาคารเรียนให้โรงเรียนการเรือนและโรงเรียนอนุบาลนครราชสีมา นายเสงี่ยม วัฒนธรรม ศึกษาธิการจังหวัดจึงสร้างในที่ราชพัสดุ ถนนอัษฎางค์ เป็นอาคาร ๒ หลัง สำหรับโรงเรียนการเรือนและโรงเรียนอนุบาลนครราชสีมา (โรงเรียนเทศบาล ๓ ในปัจจุบัน)

ได้รับคำสั่งแต่งตั้งให้ นางสาวลดาวัลย์ จิตะสมบัติ รักษาการในตำแหน่งครูใหญ่โรงเรียนอนุบาลนครราชสีมา

ได้รับงบประมาณสร้างอาคารเรียนในที่ดินราชพัสดุ (ถนนมิตรภาพ ในปัจจุบัน) บริเวณเนื้อที่ ๑๔ ไร่ ๒ งาน มีอาคารเรียน ๔ ห้อง ในขณะนั้นเอกชนเช่าทำสวนผัก

พ.ศ. ๒๔๙๗ ได้รับงบประมาณต่อเติมอาคารเรียนอีก ๕ ห้องเรียน รวมเป็น ๙ ห้องเรียน

พ.ศ. ๒๔๙๘ โรงเรียนได้ย้ายไปทำการสอนในสถานที่ๆ สร้างใหม่ ซึ่งเป็นที่ตั้งโรงเรียนในปัจจุบัน เมื่อวันที่ ๒๒ เมษายน พ.ศ. ๒๔๙๘ เปิดทำการสอนชั้นอนุบาลปีที่ ๑ ถึงประถมศึกษาปีที่ ๔ ต่อมาได้เปิดถึงชั้นประถมศึกษาปีที่ ๖

โรงเรียนได้เจริญก้าวหน้ามาเป็นลำดับ ได้รับงบประมาณ เงินบริจาคจากผู้ปกครอง ได้รับการช่วยเหลือปรับปรุงบริเวณโรงเรียน และอาคารเรียนจากหน่วยราชการต่างๆ จากเงินที่โรงเรียนจัดงานหารายได้พิเศษ นำมาก่อสร้างต่อเติมอาคารเรียน สร้างอาคารประกอบ การจัดหาพัสดุ ครุภัณฑ์ ให้มีปริมาณเพียงพอกับจำนวนนักเรียนที่เพิ่มขึ้นอย่างรวดเร็ว ประชาชนสนใจนำบุตรหลานมาฝากเข้าเรียนเป็นจำนวนมากทุกปี

โรงเรียนอนุบาลนครราชสีมาเป็นโรงเรียนขนาดใหญ่ที่สุดในประเทศไทย จะเห็นได้จากโรงเรียนต่างๆ ทั่วประเทศมาศึกษาดูงานอยู่เสมอ

พ.ศ. ๒๕๒๔ นางลดาวัลลย์ วรรณบูรณ์ ได้รับการคัดเลือกจากคณะกรรมการการประถมศึกษาแห่งชาติ ให้เป็นผูบริหารดีเด่น

พ.ศ. ๒๕๒๕ โรงเรียนอนุบาลนครราชสีมา ได้รับคัดเลือกเป็นโรงเรียนดีเด่นของคณะกรรมการการประถมศึกษาแห่งชาติ

พ.ศ. ๒๕๒๗ นางลดาวัลลย์ วรรณบูรณ์ ได้รับคัดเลือกเป็นข้าราชการบำนาญดีเด่นของข้าราชการบำนาญของคณะกรรมการการประถมศึกษาแห่งชาติ กระทรวงศึกษาธิการ

พ.ศ. ๒๕๒๗ นางสงวนศรี สร้อยสุวรรณ มาดำรงตำแหน่งผู้อำนวยการโรงเรียนในวันที่ ๖ มกราคม พ.ศ. ๒๕๒๗ ในปีนี้โรงเรียนได้รับรางวัลพระราชทานระดับประถมศึกษาขนาดใหญ่ของกระทรวงศึกษาธิการ

โรงเรียนได้รับเงินงบประมาณการก่อสร้างอาคารเรียนแบบ สปช. ๒/๒๘ รวม ๑๘ ห้องเรียน ได้ปลูกสร้างอาคารจริยศึกษาที่ประดิษฐานพระพุทธรูป ป้อมยามทรงไทย และสวนสัตว์ จัดตั้งห้องปฏิบัติการทางภาษา (Sound Lab) จำนวน ๕๐ ที่นั่ง โดยได้รับเงินสนับสนุนจากสมาคมผู้ปกครองและครูโรงเรียนอนุบาลนครราชสีมา จำนวน ๑๓๐,๐๐๐ บาท เมื่อวันที่ ๒๐ กันยายน ๒๕๒๙

พ.ศ. ๒๕๒๙ นางประภัสศรี ฉายศรีววรรณ มาดำรงตำแหน่งผู้อำนวยการโรงเรียน ในเดือนธันวาคม พ.ศ. ๒๕๒๙
