จังหวัดศรีสะเกษเริ่มขึ้นในสมัยกรุงศรีอยุธยาตอนปลาย เมื่อ พ.ศ. 2302 สมเด็จพระเจ้าเอกทัศน์โปรดให้ยกบ้านปราสาทสี่เหลี่ยมดงลำดวนขึ้นเป็นเมืองนครลำดวน ต่อมาเมืองนครลำดวนเกิดภาวะขาดแคลนน้ำจึงโปรดเกล้าฯ ให้เทครัวไปจัดตั้งเมืองใหม่ที่ริมหนองแตระห่างจากเมืองเดิมไปทางใต้ เมืองใหม่เรียก "เมืองขุขันธ์" หรือ "เมืองคูขัณฑ์" ซึ่งได้แก่อำเภอขุขันธ์ในปัจจุบัน

ต่อมาพระบาทสมเด็จพระพุทธยอดฟ้าจุฬาโลกโปรดเกล้าฯ ให้แยกบ้านโนนสามขาสระกำแพงออกจากเมืองขุขันธ์ แล้วตั้งเป็นเมืองใหม่เรียก เมืองศรีสะเกศ

ครั้นรัชสมัยพระบาทสมเด็จพระปรมินทรมหาจุฬาลงกรณ์ พระจุลจอมเกล้าเจ้าอยู่หัว ใน พ.ศ. 2455 จึงโปรดให้รวมบ้านเมืองขุขันธ์ เมืองศรีสะเกศ และเมืองเดชอุดม เข้าเป็นเมืองเดียวกันเรียก "เมืองขุขันธ์"

ซึ่งต่อมาใน พ.ศ. 2459 มีประกาศกระทรวงมหาดไทย เรื่อง ทรงพระกรุณาโปรดเกล้าฯ ให้เปลี่ยนคำว่าเมืองเรียกว่าจังหวัด ลงวันที่ 19 พฤษภาคม ปีนั้นเอง[2] เมืองขุขันธ์จึงเรียกใหม่เป็น "จังหวัดขุขันธ์" ตามนั้น

ครั้นรัชสมัยพระบาทสมเด็จพระปรเมนทรมหาอานันทมหิดล พระอัฐมรามาธิบดินทร ในวันที่ 11 พฤศจิกายน พ.ศ. 2481 คณะผู้สำเร็จราชการแทนพระองค์ได้ตราพระราชกฤษฎีกาเปลี่ยนนามจังหวัดและอำเภอบางแห่ง พุทธศักราช 2481[3] มาตรา 3 ให้เปลี่ยนชื่อ "จังหวัดขุขันธ์" เป็น จังหวัดศรีสะเกษ (เดิมในพระราชกฤษฎีกาสะกดว่า "ศีร์ษะเกษ") นับแต่นั้น ทั้งนี้ พระราชกฤษฎีกาดังกล่าวมีผลใช้บังคับตั้งแต่วันที่ 14 พฤศจิกายน พ.ศ. 2481
