พระประวัติ
พระองค์เจ้ารัชนีแจ่มจรัสเป็นพระโอรสในกรมพระราชวังบวรวิไชยชาญ (พระองค์เจ้า ยอดยิ่งยศ พระราชโอรสในพระบาทสมเด็จพระปิ่นเกล้าเจ้าอยู่หัว) กับ เจ้าจอมมารดาเลี่ยม (เล็ก) ธิดานายสุดจินดา (พลอย ชูโต) ประสูติเมื่อวันที่ 10 มกราคม พ.ศ. 2419 ในพระบวรราชวัง มีพระเชษฐภคินีหนึ่งพระองค์ คือ พระองค์เจ้าภัททาวดีศรีราชธิดา (พ.ศ. 2414 - 2442)
เมื่อเยาว์วัยเรียนหนังสือกับมารดาที่ตำหนัก เมื่อชันษา 5 ขวบ ก็ทรงอ่านหนังสือได้คล่อง ทรงเข้าศึกษาที่โรงเรียนพระตำหนักสวนกุหลาบ เมื่อ พ.ศ. 2429 และต่อมาศึกษาภาษาอังกฤษจนถึง พ.ศ. 2436 จึงเข้ารับราชการในตำแหน่งนายเวร กระทรวงธรรมการ ขณะพระชันษาได้ 16 ปี และได้เลื่อนเป็นผู้ช่วยในกรมศึกษาธิการใน ทรงรับหน้าที่พิเศษเป็นข้าหลวงสอบไล่วิชาหนังสือไทย ทรงเป็นกรรมการพิเศษร่างพระราชบัญญัติพิจารณาความแพ่ง และทรงได้เลื่อนเป็นผู้ช่วยที่ปรึกษากระทรวงพระคลังมหาสมบัติ
เมื่อพระบาทสมเด็จพระจุลจอมเกล้าเจ้าอยู่หัว เสด็จประพาสยุโรป พ.ศ. 2440 ทรงพระกรุณาโปรดเกล้าฯ ให้พระองค์เจ้ารัชนีแจ่มจรัสตามเสด็จด้วย และทรงศึกษาต่อในมหาวิทยาลัยเคมบริดจ์ เป็นเวลา 2 ปี เสด็จกลับจากประเทศอังกฤษเมื่อ พ.ศ. 2442
พระราชวรวงศ์เธอ กรมหมื่นพิทยาลงกรณ์ โปรดเล่นกีฬาเทนนิส ทรงพระดำริตั้ง ลอนเทนนิสสมาคมแห่งประเทศไทย ขึ้นเมื่อวันที่ 14 เมษายน พ.ศ. 2470
พระกรณียกิจ
พ.ศ. 2442 ทรงเข้ารับราชการในกระทรวงพระคลังมหาสมบัติ ทรงรับตำแหน่งเป็นผู้ช่วยอธิบดีกรมตรวจและกรมสารบาญชี
พ.ศ. 2444 ทรงย้ายเป็นปลัดกรมธนบัตร และเจริญก้าวหน้าเป็น ผู้แทนเจ้ากรมธนบัตร เจ้ากรมกองที่ปรึกษาอธิบดีกรมประสาปน์สิทธิการ อธิบดีกรมตรวจและกรมสารบาญชี อธิบดีกรมพาณิชย์และสถิติพยากรณ์ ทรงจัดตั้งและวางรากฐานกิจการสหกรณ์ จนในที่สุดได้เลื่อนตำแหน่งเป็นรองเสนาบดี กระทรวงพาณิชย์
พ.ศ. 2454 พระบาทสมเด็จพระมงกุฏเกล้าเจ้าอยู่หัว ทรงพระกรุณาโปรดเกล้าฯ ตั้งให้พระราชวรวงศ์เธอ พระองค์เจ้ารัชนีแจ่มจรัสเป็น องคมนตรี และโปรดเกล้าฯ ให้ทรงกรมฯ เป็น กรมหมื่นพิทยาลงกรณ์ เมื่อ พ.ศ. 2456
พ.ศ. 2468 พระบาทสมเด็จพระปกเกล้าเจ้าอยู่หัว ทรงพระกรุณาโปรดเกล้าฯ ให้พระราชวรวงศ์เธอ กรมหมื่นพิทยาลงกรณ์ ดำรงตำแหน่งอุปนายกกรรมการหอพระสมุดสำหรับพระนคร ซึ่งต่อมาได้รวมเข้ากับกรมศิลปากร ต่อมาได้เปลี่ยนจากหอสมุดสำหรับพระนครเป็น "ราชบัณฑิตยสภา"
