พิธีวันมหิดลเริ่มโดยสโมสรนักศึกษาแพทย์ศิริราช ภายหลังจากที่พระบาทสมเด็จพระเจ้าอยู่หัวภูมิพลอดุลยเดช เสด็จฯทรงเปิด พระบรมรูปสมเด็จพระราชบิดา เมื่อวันที่ 27 เมษายน พ.ศ.2493 แล้ว ในวันที่ 24 กันยายน ปีเดียวกัน สโมสรนักศึกษาแพทย์ศิริราช และนักศึกษาพยาบาล โดยการนำของ นายบุญเริ่ม สิงหเนตร นายกสโมสรนักศึกษาแพทย์ และ นางสาวชายัญ ปรักกะมะกุล หัวหน้านักศึกษาพยาบาล นำนักศึกษาแพทย์และนักศึกษาพยาบาลตั้งแถวตามถนนจักรพงษ์ หน้าพระบรมรูปสมเด็จพระราชบิดา จากนั้นหัวหน้านักศึกษาวางพวงมาลาแล้วผู้แทนนักศึกษาอ่านฉันท์ ทูลกระหม่อมสดุดีอศิรวาท ซึ่งประพันธ์โดย นายภูเก็ต วาจานนท์ หลังจากนั้น ศ.นพ.สุด แสงวิเชียร และ ศ.นพ.เติม บุนนาค วางพวงมาลาของคณะแพทยศาสตร์ศิริราชพยาบาล
