อาจารย์มนตรี ตราโมท เดิมชื่อ บุญธรรม ตราโมท เกิดวันที่ 17 มิถุนายน พ.ศ. 2443 ที่บ้านท่าพี่เลี้ยง อำเภอเมือง จังหวัดสุพรรณบุรี เป็นบุตรนายยิ้ม และนางทองอยู่ สมรสกับนางสาวลิ้นจี่
(บุรานนท์) เมื่อ พ.ศ. 2475 มีบุตร 2 คน คือ นายฤทธี และนายศิลป์ ต่อมาเมื่อนางลิ้นจี่ ถึงแก่กรรมจึงแต่งงานกับนางสาวพูนทรัพย์ (นาฏประเสริฐ) มีบุตร 2 คน คือ นางสาวดนตรี และนายญาณี

อาจารย์มนตรี ตราโมท เริ่มการศึกษา ที่โรงเรียนประจำจังหวัดสุพรรณบุรี

ได้รู้จักกับนักดนตรีในวงและขอเข้าไปเล่นด้วย เมื่อมีการบรรเลงก็มักจะไปช่วยตีฆ้องเล็กหรือทุ้มเหล็กด้วยเสมอเมื่อจบมัธยมศึกษาปีที่ 3 ครูสมบุญ นักฆ้องซึ่งเป็นครูปี่พาทย์ประจำวง ชวนให้มาหัดปี่พาทย์จึงได้เรียนอย่างจริงจังประมาณ 2 ปี และได้เป็นนักดนตรีประจำวงปี่พาทย์ ต่อมา ได้ไปศึกษาเพิ่มเติมด้านปี่พาทย์ที่จังหวัดสมุทรสาคร ราว พ.ศ. 2456 ที่บ้านครูสมบุญ สมสุวรรณ ที่บ้านนี้มีวงปี่พาทย์และแตรวง จึงได้ฝึกทั้ง 2 อย่างคือ ด้านปี่พาทย์ ฝึกระนาดเอกและฆ้องวงใหญ่ ด้านแตรวง ฝึกเป่าคลาริเน็ต นอกจากนี้
ครูสมบุญยังได้แนะวิธีแต่งเพลงให้ด้วย
ในปี พ.ศ. 2460 ได้เข้ามาสมัครรับราชการในกรมพิณพาทย์หลวง ซึ่งมีพระยาประสาน
