ปี พ.ศ. 2443 เจ้าพระยาพระเสด็จสุเรนทราธิบดี เสนาบดีกระทรวงธรรมการได้จัดการเรียนการสอนระดับมัธยมศึกษาและสร้างอาคารเรียน นับเป็นโรงเรียนสายสามัญโรงเรียนแรกของจังหวัดสมุทรปราการต่อมาได้กลายเป็น โรงเรียนสมุทรปราการ
พ.ศ. 2445 พระยาราชายุสาธก นายด่านภาษี ได้เพิ่มอาคารเรียนสำหรับผู้หญิง ชื่อโรงเรียนสตรีสมุทรปราการและต่อมา มีเด็กชายเข้ามาเรียนด้วย กลายเป็น โรงเรียนสหศึกษา

ปี พ.ศ. 2481 โรงเรียนสตรีสมุทรปราการ แยกโอนนักเรียนหญิงมาจากโรงเรียน สมุทรปราการและตั้งโรงเรียนขึ้น ณ บริเวณวัดพิชัยสงครามบนเนื้อที่ประมาณ 3 ไร่ เศษ มีนักเรียนชั้นป. 3 ถึงม.3 ครูใหญ่คนแรคือ นางกระแสร์ รัตนวราหะ
ปี พ.ศ. 2485 นางสาวสบสมัย บุราวาศ ครูใหญ่โรงเรียนสตรีอุทัยธานี ย้ายมาดำรง ตำแหน่งแทน โรงเรียนสตรีสมุทรปราการมีความเจริญก้าวหน้าเพิ่มมากขึ้น มีผู้จ้องการเข้ามาศึกษาเพิ่มขึ้นมากจนต้องขยายโรงเรียน เพราะเดิมคับแคบ
วันที่ 8 สิงหาคม พ.ศ. 2499 โรงเรียนสตรีสมุทรปราการได้ย้ายจาก สถานที่เดิมไปตั้งอยู่ ณ เลขที่ 489 ถนนสุขุมวิท ตำบลปากน้ำ อำเภอเมืองฯ จังหวัดสมุทรปราการ ปัจจุบันรวมเนื้อที่ของโรงเรียน ทั้งหมด 11 ไร่ 47 ตารางวา
ปัจจุบันโรงเรียนสตรีสมุทรปราการ เป็นโรงเรียนมัธยมขนาดใหญ่พิเศษ แบบสหศึกษา สังกัดสำนักงานเขตพื้นที่การศึกษามัธยมศึกษา เขต 6 เปิดทำการสอน ตั้งแต่ชั้นมัธยมศึกษาปีที่ 1-6
