During the fermentation 150 ml samples were taken every 4 h
for analysis. Cells number in fermentation medium was
determined using a Neubauer Haemocytometer and a microscope
at 400 magnification. Viability was determined using
methylene blue staining technique. The free amino nitrogen,
pH and conductivity were determined on laboratory multiparameter
analyser Consort C863 (Consort, Belgium). The
samples were centrifuged at 4000 rpm for 15 min, and the
fermentable sugar content (sucrose, glucose and fructose) of
the supernate was determined by HPLC (Jasco, pump PU-980,
detector RI-930, sampler AS-950, 20 ml injection loop, column
Sugar KS-801, eluent: water at a flow rate of 0.6 ml min1 and
elution time 30 min). CO2 weight loss in fermentation was
measured by an accurate balance (0.01 g).
