เขตรักษาพันธุ์สัตว์ป่าทุ่งใหญ่นเรศวร ตั้งอยู่ในท้องที่ตำบลไล่โว่ อำเภอสังขละบุรี จังหวัดกาญจนบุรี และตำบลแม่ละมุ้ง อำเภออุ้มผาง จังหวัดตาก มีเนื้อที่ทั้งหมดประมาณ ๓,๒๐๐ ตารางกิโลเมตร หรือประมาณ ๒ ล้านไร่ โดยมีอาณาเขตติดต่อดังนี้
