วัดธรรมิกราช เดิมชื่อ วัดมุขราช ตั้งอยู่ใน อำเภอพระนครศรีอยุธยา จังหวัดพระนครศรีอยุธยา ติดกับพระราชวังโบราณ และวัดพระศรีสรรเพชญ์ ปัจจุบันยังเป็นวัดที่มีพระภิกษุสงฆ์จำพรรษาและปฏิบัติธรรมอยู่ โดยมีพระครูสมุห์ธรรมภณเป็นเจ้าอาวาส
เมื่อพระเจ้าสายน้ำผึ้ง สร้างวัดพนัญเชิงนั้น พระราชโอรส คือพระเจ้าธรรมิกราช โปรดให้สร้างวัดนี้ขึ้นที่บริเวณเมืองเก่าชื่อเมืองอโยธยาศรีรามเทพนคร ทางหน้าประตูด้านทิศเหนือคือ พระเจดีย์สิงห์ล้อม ๕๒ ตัวที่แตกต่างไปจากเจดีย์ทั่วไป นับเป็นพระเจดีย์สิงห์แห่งเดียวในพระนครศรีอยุธยา ก่อนสร้างกรุงศรีอยุธยา ในสมัยต่อมา พระมหากษัตริยได้ทรงบูรณะมาโดยตลอด โดยสังเกตจากร่องรอยการซ่อม และพื้นที่ของวัดที่อยู่ทางทิศตะวันออกของพระนครฯ ตามคติโบราณถือว่าเป็นทิศมงคล ในสมัยสมเด็จพระไตรโลกนาถทรงธรรม (พ.ศ. ๒๑๕๓) ทรงบูรณะวัด และสร้างพระวิหารหลวง เพื่อฟังธรรมในวันธรรมสวนะ (วันพระ) สำหรับพระวิหารพระพุทธไสยยาสน์ (พระนอน) นั้น พระราชมเหสีของพระองค์มีพระราชธิดาประชวร ทรงอธิษฐานไว้เมื่อพระราชธิดาหายแล้วจึงสร้างพระวิหารถวาย น้ำพระพุทธมนต์ในพระวิหารนี้กล่าวกันว่ามี ความศักดิ์สิทธิ์มากมีประชาชนมาอธิษฐาน ขอไปใช้ตามความปรารถนาจำนวนมาก
