ร้อยโทหญิงปวีณา ทองสุก เกิดที่ อำเภอศีขรภูมิ จังหวัดสุรินทร์ เป็นลูกคนที่ 2 ของนายนิรันดร์-นางสุกรี ทองสุก จบการศึกษาชั้นประถมศึกษา จากโรงเรียนบ้านสว่าง ระดับมัธยมศึกษาตอนต้น ที่โรงเรียนศรีขรภูมิ จังหวัดสุรินทร์ และมัธยมศึกษาตอนปลาย จากโรงเรียนเทพลีลา บางกะปิ กรุงเทพฯ ได้รับการชักชวนให้มาเล่นกีฬายกน้ำหนัก ขณะยังเรียนอยู่ชั้น ม.2 โดยมี อาจารย์วินัย คำจีนศรี เป็นผู้ฝึกสอน เข้าแข่งขันระดับนานาชาติครั้งแรก ในเอเชียนเกมส์ 1998 ที่กรุงเทพมหานคร แต่ไม่ได้รับเหรียญรางวัล.

การแข่งขัน โอลิมปิกฤดูร้อน 2004 ที่ เอเธนส์ ประเทศกรีซ วันที่ 21 สิงหาคม พ.ศ. 2547 ร้อยโทหญิงปวีณา ลงแข่งขันในรุ่นน้ำหนัก 75 กิโลกรัม ท่าแรก คือท่าสแนทช์ ยกได้ 122.5 ก.ก. เป็นอันดับสองรองจากวาเลติน่า เลโปว่า จากประเทศรัสเซีย ซึ่งยกได้ 125 ก.ก. ท่าที่สอง คือคลีนแอนด์เจิร์ก ปวีณายกได้ 150 ก.ก. ซึ่งเป็นสถิติโอลิมปิก ในขณะที่วาเลติน่ายกได้เพียง 147.5 ก.ก. จึงมีน้ำหนักรวมเท่ากัน แต่นางสาวปวีณา มีน้ำหนักตัวน้อยกว่า จึงได้เหรียญทองไปครอง ถือเป็นนักกีฬาหญิงคนที่สองของไทย ถัดจาก อุดมพร พลศักดิ์ ซึ่งได้เหรียญจากรุ่น 53 ก.ก. ไปก่อนแล้ว

นางสาวปวีณา ได้รับเลือกจากสหพันธ์ยกน้ำหนักนานาชาติให้เป็นนักกีฬายกน้ำหนักหญิงยอดเยี่ยม ของการแข่งขันโอลิมปิกเกมส์ครั้งดังกล่าว นอกจากนี้ มหาวิทยาลัยราชภัฏสุรินทร์ ได้มอบปริญญาวิทยาศาสตรดุษฎีบัณฑิตกิตติมศักดิ์ โปรแกรมวิชาวิทยาศาสตร์การกีฬาให้ ในพิธีมอบปริญญา ประจำปี พ.ศ. 2547

การแข่งขัน เอเชียนเกมส์ 2006 ที่กรุงโดฮา ประเทศกาตาร์ วันที่ 4 ธันวาคม พ.ศ. 2549 นางสาวปวีณา ลงแข่งขันในรุ่นน้ำหนัก 63 ก.ก. ในท่าสแนทช์ ยกได้ 110 ก.ก. โดยตามหลัง โอ หยาง เสี่ยว ฟาง จากประเทศจีน ที่ยกได้ถึง 115 ก.ก. ในท่าคลีนแอนด์เจิร์ก นางสาวปวีณา และเสี่ยวฟาง ยกครั้งแรก 132 ก.ก. ผ่านทั้งคู่ ก่อนที่ เสี่ยวฟาง จะยกครั้งที่สอง ที่ 137 ก.ก. ไม่ผ่าน และบาดเจ็บ จนต้องถอนตัว นางสาวปวีณา ที่ยกน้ำหนัก 137 ก.ก. ผ่าน จึงได้เหรียญทอง ก่อนจะยกครั้งที่สาม ที่ 142 ก.ก. ทำลายสถิติโลกของ สเวตลานา ชิมโคว่า
