ต่อมาในระหว่าง พ.ศ. ๒๔๓๑ - ๒๔๓๖ พระบาทสมเด็จพระจุลจอมเกล้าเจ้าอยู่หัว รัชกาลที่ ๕ ทรงให้คัดลอกพระไตรปิฎกอักษรขอมในใบลานออกมาเป็นตัวอักษรไทย แล้วชำระแก้ไขและพิมพ์ขึ้นเป็นเล่มหนังสือได้ ๔๙ เล่ม นับเป็นครั้งแรกในประเทศไทยที่ได้มีการจัดพิมพ์พระไตรปิฎกเป็นรูปเล่มหนังสือด้วยอักษรไทย
