โรงเรียนเพชรพิทยาคม ในอดีตเป็นโรงเรียนประจำมณฑลเพชรบูรณ์ ก่อตั้งขึ้นเมื่อปี พ.ศ. 2444 โดยพระยาเพชรรัตน์สงคราม ที่บริเวณวัดไตรภูมิ ตำบลในเมือง จังหวัดเพชรบูรณ์ ซึ่ง กระทรวงธรรมการได้แต่งตั้งให้นายเปล่ง เพชระบูรณิน มาดำรงตำแหน่งครูใหญ่ แรกเปิดสอนในระดับประถมศึกษา ต่อมาได้มีการย้ายสถานที่ตั้งโรงเรียนหลายครั้ง เช่น บริเวณวัดใหม่ บริเวณโรงเรียนเมืองเพชรบูรณ์ ได้เปิดสอนระดับชั้นมัธยมศึกษาตอนต้น ปี พ.ศ. 2482 และมัธยมศึกษาตอนปลาย ปี พ.ศ. 2505 จนกระทั่งปี พ.ศ. 2518 ได้ย้ายสถานที่ตั้งมาที่บริเวณโรงเรียนเพชรพิทยาคมปัจจุบันและกรมสามัญศึกษาได้แต่งตั้งนายจักรกฤษณ์ นาคะรัตน์ เป็นผู้อำนวยการคนแรก ปัจจุบัน นายพีรพัฒน์ วัชรินทรางกูร ผู้อำนวยการโรงเรียนเพชรพิทยาคม (คนที่ 31 ) ได้ย้ายมาดำรงตำแหน่ง เมื่อวันที่ 12 ธันวาคม 2555
