นที่ 3 กรกฎาคม พ.ศ. 2421 ทรงพระกรุณาโปรดเกล้าฯ อาราธนา พระอริยมุนี มาจากวัดบวรนิเวศ พร้อมด้วย พระฐานานุกรม 3 รูป พระอันดับ 16 รูป สามเณร 3 รูป รวม 23 รูป มาอยู่วัดนี้ พระบาทสมเด็จพระจุลจอมเกล้า เจ้าอยู่หัวทรงบำเพ็ญพระราชกุศล แล้วประกาศ พระราชทานวิสุงคามสีมา และ พระราชทานนามอารามแห่งนี้ว่า วัดเทพศิรินทราวาส ตามพระนามแห่งองค์พระราชชนนี

สถาปัตยกรรมสำคัญได้แก่ พระอุโบสถของวัดเทพศิรินทราวาส มีขนาดใหญ่และตกแต่งอย่างงดงาม ด้วยลายรดน้ำและซุ้มประตูหน้าต่าง เพดานสลักรูปเครื่องราชอิสริยาภรณ์ พระอุโบสถเป็นที่ประดิษฐานพระพุทธรูปสำคัญหลายองค์ เช่น พระนิรันตรายซึ่งเป็นพระพุทธรูปประจำรัชกาลที่ 4 ซึ่งอันเชิญมาเมื่อ พ.ศ. 2421 ตามพระราชประสงค์รัชกาลที่ 4
