ตั้งอยู่ที่ 110 หมู่ 5 ถ.สันติสุข ต.จันทนิมิต อ.เมือง จ.จันทบุรี มีแม่น้ำจันทบุรีไหลผ่านบริเวณด้านหน้า เป็นโบสถ์ที่มีประวัติการก่อสร้างยาวนานถึง ๓๐๐ ปี ครั้งแรกสร้างขึ้นในปี พ.ศ. ๒๒๕๔ บนฝั่งตะวันตกของแม่น้ำจันทบุรี โดยคุณพ่อเฮิ้ต โตแลนติโน และบรรดาคาทอลิกชาวญวน จนถึงปี พ.ศ. ๒๓๗๗ ได้มีการย้ายมาสร้างบนฝั่งตะวันออกของแม่น้ำจันทบุรีอันเป็นสถานที่ตั้งในปัจจุบัน โดยมิได้มีการบันทึกถึงสาเหตุของการย้ายแต่ประการใด และในปี พ.ศ. ๒๔๔๖ ได้ก่อสร้างโบสถ์หลังปัจจุบันขึ้นให้มีขนาดใหญ่กว่าหลังเก่าเพื่อรองรับกับจำนวนคริสตศาสนิกชนที่เพิ่มมากขึ้น

พ.ศ. 2528 ได้มีการจัดงานฉลองโบสถ์คาทอลิกจันทบุรีครบรอบ 75 ปี นับได้ว่าโบสถ์คาทอลิกแห่งนี้เป็นโบสถ์ขนาดใหญ่ที่มีความเก่าแก่และกล่าวกันว่ามีความงดงามมากที่สุดในประเทศ
