สำหรับการเดินทางไปยังอุทยานแห่งชาติเขาใหญ่นั้น สามารถเดินทางได้ 2 เส้นทาง คือ

- เส้นทางที่ 1
จากกรุงเทพฯ ถนนพหลโยธิน ผ่านรังสิต ประตูน้ำพระอินทร์ สระบุรี เลี้ยวขวาเข้าทางหลวงหมายเลข 2 หรือถนนมิตรภาพ ก่อนถึงอำเภอปากช่อง บริเวณ กม. 56 ชิดซ้ายขึ้นสะพานลอยไปตามถนนธนะรัชต์ทางหลวงหมายเลข 2090 ถึง กม. 23 จะพบด่านศาลเจ้าพ่อเขาใหญ่ เดินทางต่อไปอีก 15 กิโลเมตร ก็จะถึงที่ทำการอุทยานฯ รวมระยะทางประมาณ 205 กิโลเมตร

- เส้นทางที่ 2
จากกรุงเทพฯ ถนนพหลโยธิน เลี้ยวขวาบริเวณรังสิต เข้าทางหลวงหมายเลข 305 สู่ตัวเมืองนครนายก แล้วเข้าทางหลวงหมายเลข 33 (ถนนสุวรรณศร) ถึงสี่แยกศาลนเรศวรเลี้ยวซ้ายตามเส้นทางปราจีนบุรี - เขาใหญ่ ผ่านด่านตรวจค่าธรรมเนียมบริเวณเชิงเขาแล้วเดินทางต่อ จนถึงที่ทำการอุทยานแห่งชาติ รวมระยะทางประมาณ 190 กิโลเมตร
