โรงเรียนวัดสระพังลานตั้งอยู่ หมู่ที่ 6 ตำบลสระพังลาน อำเภออู่ทอง จังหวัดสุพรรณบุรี เปิดเป็นโรงเรียนประชาบาล หมู่ที่ 1 ตำบลสระยายโสม โดยใช้ศาลาชั่วคราวของวัดสระพังลานเป็นที่เรียน เมื่อวันที่ 24 กันยายน พ.ศ. 2482 สอนตั้งแต่ชั้นมูลศึกษาถึงชั้นประถมศึกษาปีที่ 4 วันที่ 1 มิถุนายน พ.ศ. 2505 ได้มาเรียนที่อาคารสร้างใหม่เป็นอาคารเรียนแบบ 012 ขนาด 5 ห้องเรียน ตึกชั้นเดียว ปี พ.ศ. 2516 เปิดสอนระดับ ป.ปลาย ในชั้นประถมศึกษาปีที่ 5 พ.ศ. 2533 ขยายการศึกษาภาคบังคับถึงชั้นประถมศึกษาปีที่ 6 พ.ศ. 2539 เปิดสอนชั้นเด็กเล็ก พ.ศ. 2535 ได้รับอนุญาตให้เปิดเป็นโรงเรียนขยายโอกาสเปิดสอนตั้งแต่ชั้นเด็กเล็กจนถึงชั้นมัธยมศึกษาปีที่ 3
