ต่อมาในสมัยนายธานินทร์ กรัยวิเชียร เป็นนายกรัฐมนตรีรัฐบาลได้เล็งเห็นว่า การปราบปรามยาเสพติดไม่สามารถแก้ไขได้โดยการดำเนินการเฉพาะกรมตำรวจฝ่ายเดียว จึงได้เสนอร่างพระราชบัญญัติป้องกันและปราบปรามยาเสพติด พ.ศ. 2519 ต่อสภาปฏิรูปการปกครองแผ่นดิน และประกาศใช้เป็นกฎหมายเมื่อวันที่ 16 พฤศจิกายน 2519


ตั้งแต่นั้นเป็นต้นมา การแก้ไขปัญหายาเสพติดของประเทศไทยก็ได้ดำเนินไปอย่างมีแบบแผนและเป็นระบบที่ดีขึ้น พระราชบัญญัติดังกล่าวได้กำหนดให้มี "คณะกรรมการป้องกันและปราบปรามยาเสพติด" หรือเรียกชื่อย่อว่า ป.ป.ส. โดยมีนายกรัฐมนตรีเป็นประธาน และจัดตั้งสำนักงาน ป.ป.ส. ขึ้นเป็นหน่วยงานกลางรับผิดชอบโดยตรง มีฐานะเป็นกรม กรมหนึ่งในสำนักนายกรัฐมนตรี มีหน้าที่ประสานงานกับหน่วยงานที่เกี่ยวข้องกับการป้องกันและปราบปรามยาเสพติดทั้งในประเทศและต่างประเทศ


สำนักงานป.ป.ส. ในฐานะหน่วยงานกลางที่รับผิดชอบเกี่ยวกับการป้องกันและแก้ไขปัญหายาเสพติดในประเทศมาโดยตลอด และได้นำมติเรื่องวันต่อต้านยาเสพติดขององค์การสหประชาชาติเสนอต่อคณะรัฐมนตรีในการประชุมเมื่อวันที่ 14 มิถุนายน 2531 ซึ่งที่ประชุมคณะรัฐมนตรีได้มีมติให้กำหนดวันที่ 26 มิถุนายนของทุกปี เป็นวันต่อต้านยาเสพติดโดยเริ่มตั้งแต่ปี พ.ศ. 2531 เป็นต้นมา
