เปิดทำการสอนเป็นโรงเรียนพาณิชยการ จากอาคารไม้เก่าแก่ซึ่งมีอยู่เป็นเวลาถึง 90 ปี แต่เดิมเป็นโรงเรียนอาชีวพาณิชยการวัดมหาพฤฒาราม รวมเวลา 30 ปี ( ปัจจุบันเป็นโรงเรียนสารพัดช่าง ซึ่งย้ายจากวัดมหาพฤฒารามไปอยู่ที่วัดแก้วแจ่มฟ้า )
ต่อมาเมื่อ พ.ศ. 2461 กระทรวงศึกษาธิการให้งบประมาณ 5,000 บาท เพื่อทำการซ่อมใหม่แล้วเปลี่ยนชื่อเป็นโรงเรียนสตรีวัดมหาพฤฒาราม ซึ่งขึ้นตรงต่อกรมสามัญศึกษา ( เดิม ) เริ่มรับสมัครนักเรียนเมื่อวันที่ 3 ธันวาคม พ.ศ. 2461 และเปิดทำการสอนครั้งแรกเมื่อวันที่ 12 ธ.ค. 2461 โดยรับนักเรียนที่มีอายุ 6 ขวบ ขึ้นไป ทั้งชายและหญิง นักเรียนชายให้เรียนอยู่ได้อายุไม่เกิน 12 ปี สอนตั้งแต่ชั้นประถมศึกษาปีที่ 1 ถึงชั้นประถมศึกษาปีที่ 3 มีจำนวนนักเรียน 109 คน ครู 3 คน นางสุภาพ ตีระนันทน์ เป็นครูใหญ่ ต่อมาได้เพิ่มชั้นเรียนตามลำดับ จากชั้นประถมศึกษาปีที่ 3 จนถึงมัธยมปีที่ 8 พ.ศ. 2482 ยุบชั้นมัธยมปีที่ 7 –8 ตามแผนการศึกษาของชาติสมัยนั้น
พ.ศ. 2544 ได้เร่งพัฒนาทางด้านวิชาการ พร้อมกับการพัฒนาของโรงเรียนเพื่อให้โรงเรียนมีความพร้อมเต็มที่ในการปฏิรูปการศึกษา โดยเร่งส่งเสริมให้มีแหล่งเรียนรู้ สภาพแวดล้อมที่เอื้อต่อการเรียนการสอนสนับสนุนให้มีครูแกนนำ ครูต้นแบบ ครูแห่งชาติ ฯลฯ ตลอดจนการระดมสรรพกำลังจากชุมชนท้องถิ่น เพื่อประโยชน์ต่อการพัฒนาการศึกษาของโรงเรียน ทั้งนี้โรงเรียนมีการเน้นการพัฒนานักเรียนที่มีความสามารถพิเศษเฉพาะด้าน และพัฒนาครูอาจารย์ด้านทักษะการจัดกระบวนการเรียนรู้ที่ฝึกให้นักเรียนคิดและปฏิบัติตามยุทธศาสตร์การปฏิรูปการศึกษา
