โรงเรียนแก่นนครวิทยาลัย จังหวัดขอนแก่น เริ่มก่อตั้งเมือเดือนพฤษภาคม พ.ศ 2510 โดยมี อาจารย์เจือ หมายเจริญ ซึ่งเป็นอาจารย์ใหญ่โรงเรียนขอนแก่นวิทยายนและรักษาการในตําแหน่งครูใหญ่ โรงเรียนแก่นนครวิทยาลัยอีกตําแหน่งหนึ่ง ในปีแรกเปิดสอน 8 ห้องเรียน มีนักเรียน 280 คน แบ่งเป็นนักเรียนชาย 140 คน และนักเรียนหญิง 140 คน เป็นโรงเรียนัธยมแบบประสมและเป็นสหศึกษาคือรับทั้งชายทั้งหญิงมาเรียนรวมกัน โดยในสมัยนั้น การจัดการศึกษาในระดับมัธยมศึกษาจะแยกนักเรียนชายให้เรียนในโรงเรียนประจำ จังหวัดชาย (โรงเรียนขอนแก่นวิทยายน), นักเรียนหญิงเรียนในโรงเรียนประจำจังหวัดหญิง (โรงเรียนกัลยาณวัตร) โรงเรียนสหศึกษาคือมีนักเรียนชายและหญิงเรียนรวมกัน ส่วนโรงเรียนมัธยมแบบประสม หมายถึง การเปิดสอนในหลักสูตรที่กว้าง หลากหลาย มีวิชาให้เลือกเรียนได้ตามความรู้ความสามารถที่ตนถนัด (TO EACH HIS OWN ABILITY) มีทั้งวิชาสามัญ วิชาศิลป-ปฏิบัติ(วิชาชีพ) เมื่อจบมัธยมต้นแล้วสามารถจะนำไปศึกษาต่อหรือประกอบอาชีพได้

ในปีแรกที่เปิดการสอนต้องอาศัยโรงเรียนขอนแก่นวิทายนเรียนไปก่อนพร้อมๆกับ โรงเรียนก็เริ่มก่อสร้างไปด้วยจนถึงวันที่ 16 กันยายน 2511 จึงได้ย้ายมาเรียน ณ สถานที่ปัจจุบัน จึงถือเอาวันนี้เป็นวันเกิดของโรงเรียนด้วย

เดิมโรงเรียนนี้มีพื้นที่ 32 ไร่เศษอยู่ห่างจากตัวเมืองประมาณ 3 กิโลเมตร ใน พ.ศ. 2518 โรงเรียนได้ที่ดินราชพัสดุซึ่งเป็นสนามกีฬาจังหวัด ซึ่งอยู่ติดกับ โรงเรียน อีกหกไร่ ในปี พ.ศ. 2523 โรงเรียนได้ขอใช้ที่สนามกีฬาอีก 25 ไร่ รวมพื้นที่ทั้งหมดเป์น 63 ไร่ 1 งาน 37 ตารางวา และอีกส่วนหนึ่งเป็นบ้านพักครูมีเนื้อที่ 17 ไร่ โรงเรียนแก่นนครวิทยาลัย ได้รับพระราชทานเกียรติบัตรโรงเรียนดีเด่นขนาดใหญ่ ของเขตการศึกษา 9 ประจำปีการศึกษา 2525 เป็นโรงเรียนแรกของจังหวัดขอนแก่น
