คณะกรรมการการปิโตรเลียมแห่งประเทศไทย (ปตท.) ได้อนุมัติให้ ปตท. จัดตั้ง "ศูนย์วิจัยและพัฒนา" ขึ้นเมื่อวันที่ 13 มกราคม 2536 ณ ศูนย์ปฏิบัติการระบบท่อเขต 2 อำเภอวังน้อย จังหวัดพระนครศรีอยุธยา เพื่อที่จะพัฒนาให้เป็นศูนย์วิจัย และพัฒนาระดับประเทศ แห่งแรก ที่มีขีดความสามารถสูงเฉพาะทางในสาขาปิโตรเลียม และปิโตรเคมี ด้วยงบประมาณการก่อสร้างประมาณ 1,463 ล้านบาท
การก่อสร้างศูนย์วิจัยและพัฒนา ณ อำเภอวังน้อย จังหวัดพระนครศรีอยุธยานี้ ในส่วนของการออกแบบทาง Conceptual Designได้ให้ Southwest Research Institute ประเทศสหรัฐอเมริกาออกแบบห้องปฏิบัติการทดสอบทางเครื่องยนต์ และ BEICIP FRANLAB ประเทศฝรั่งเศสออกแบบห้องปฏิบัติการทดสอบทางเคมีและศูนย์ฝึกอบรม นอกจากนี้การดำเนินการศึกษาวางแผนแม่บท ได้ให้บริษัท ENIRICERCHE ประเทศอิตาลี
ซึ่งมีประสบการณ์ด้านการวิจัยกว่า 30 ปี เป็นผู้ศึกษา สำหรับโครงสร้างของศูนย์วิจัยและพัฒนานั้น ได้รับความเห็นชอบจากคณะกรรมการ ปตท. ในการปรับปรุงโครงสร้างและเปลี่ยนชื่อศูนย์วิจัยและพัฒนา ปตท. เป็นสถาบันวิจัยและเทคโนโลยี ปตท. เมื่อวันที่ 7 พฤศจิกายน 2540 ทั้งนี้เพราะศูนย์วิจัย ปตท.ได้ขยายขอบเขตความรับผิดชอบจากการวิจัยไปถึงการติดตามและคิดค้นเทคโนโลยีใหม่ๆ เพื่อให้ ปตท. มีขีดความสามารถใน
การพัฒนาผลิตภัณฑ์ที่มีคุณภาพดี ราคาประหยัด ทำให้สามารถแข่งขันในตลาดได้ เพราะการพึ่งพาเทคโนโลยีที่มีจำหน่ายนั้น ย่อมไม่สามารถสร้างความโดดเด่นให้ผลิตภัณฑ์ได้ ในการนี้ สถาบันวิจัยฯ ได้ส่งเสริมการดำเนินธุรกิจของ ปตท. ให้ประสบความสำเร็จ
พื้นที่สถาบันวิจัยและเทคโนโลยี ปตท. ณ อ.วังน้อย จ.พระนครศรีอยุธยา ตั้งแต่เริ่มก่อตั้ง มีทั้งสิ้น 75 ไร่ โดย 52 ไร่ ใช้ประโยชน์เป็นทั้งอาคารสำนักงาน อาคารห้องปฏิบัติการ โรงงานนำร่อง Bio-Hydrogenated Diesel โรงงานนำร่องสำหรับการทดลองกระบวนการ Hydroprocessing อาคาร Utilities โดยห้องปฏิบัติการ สถาบันวิจัยและเทคโนโลยี ปตท. อันทันสมัย ประกอบด้วย
