เริ่มการศึกษาชั้นประถมศึกษาที่โรงเรียนวัดจักรวรรดิราชาวาส (วัดสามปลื้ม) และย้ายไปศึกษาต่อที่โรงเรียนสุขุมาลวิทยาลัย จนกระทั่งเข้าศึกษาต่อที่โรงเรียนนายร้อยทหารบก ต่อมาเมื่ออายุ ๑๖ ปี ได้รับทุนรัฐบาลไปศึกษาต่อที่โรงเรียนนายร้อยทหารบกในเมืองโกรสลิสเตอร์ เฟล เด ประเทศ เยอรมันนี ศึกษาอยู่ ๓ ปี ต่อจากนั้นได้เข้าประจำอยู่ในกองทัพบกเยอรมัน สังกัดกรมทหารปืนใหญ่ที่ ๔ ในปี พ.ศ.๒๔๕๕ ได้เดินทางไปศึกษาต่อวิชาช่างแสงที่ประเทศเดนมาร์ก เรียนได้ปีเดียวก็ถูกเรียกตัวกลับเนื่องจากเงินทุนการศึกษาไม่เพียงพอ

พระยาพหลพลพยุหเสนาเข้ารับราชการครั้งแรกประจำกรมทหารปืนใหญ่ที่ ๔ จังหวัดราชบุรี อีกสามปีต่อมาได้เข้าประจำกรมทหารปืนใหญ่ บางซื่อ พระนคร และในปี พ.ศ.๒๔๖๐ ได้ย้ายไปเป็นผู้บังคับการกรมทหารปืนใหญ่ที่ ๙ จังหวัดฉะเชิงเทรา ในชีวิตราชการนั้นได้รับพระราชทานบรรดาศักดิ์เป็นหลวงและพระตามลำดับในราชทินนามเดียวกันว่า " สรายุทธสรสิทธิ์" และได้ เลื่อนยศทางทหารมาตามลำดับ กระทั่งได้เป็น พันเอก เมื่อวันที่ ๑ เมษายน ๒๔๗๑
เมื่อวันที่ ๒๐ พฤษภาคม ๒๔๗๑ มีพระบรมราชโองการโปรดเกล้าฯ ให้เป็นองครักษ์เวร และเมื่อวันที่ ๖ พฤศจิกายน ๒๔๗๔ ทรงพระกรุณาโปรดเกล้าฯ เลื่อนบรรดาศักดิ์เป็นพระยาพหลพลพยุหเสนา มีราชทินนามเดียวกับบิดา
เมื่อวันที่ ๒๔ มิถุนายน ๒๔๗๕ ได้ร่วมกับคณะราษฏรโดยเป็นหัวหน้าคณะราษฎรทำการ ยึดอำนาจเปลี่ยนแปลงการปกครอง จากระบอบสมบูรณาญาสิทธิราชย์ มาเป็นระบอบประชาธิปไตย มีรัฐธรรมนูญเป็นกฎหมายสูงสุด
พันเอก พระยาพหลพลพยุหเสนา ดำรงตำแหน่งนายกรัฐมนตรีเมื่อวันที่ ๒๑ มิถุนายน ๒๔๗๖ โดยการทำรัฐประหารรัฐบาลของพระยามโนปกรณ์นิติธาดาตลอดระยะเวลาที่บริหารประเทศ ต้องเผชิญปัญหานานัปการ จนต้องลาออกจากตำแหน่งหลายครั้ง และได้มีพระบรมราชโองการโปรดเกล้าฯ แต่งตั้งให้กลับเข้ามาดำรงตำแหน่งใหม่ ในที่สุดเมื่อวันที่ ๑๑ กันยายน ๒๔๘๑ พระยาพหลพลพยุหเสนา ได้ตัดสินใจยุบสภาและลาออกจากตำแหน่งนายกรัฐมนตรี แต่ถึงแม้พันเอก พระยาพหลพลพยุหเสนาจะวางมือจากตำแหน่งสูงสุดทางการเมืองแล้ว ในระหว่างสงครามโลกครั้งที่ ๒ คนได้ดำรงตำแหน่งแม่ทัพใหญ่ และได้รับยศ พลเอก พระยาพหลพลพยุหเสนา
