ในปี พ.ศ. ๒๔๗๓ พระครูพิศาลอรัญญเขต (จันทร์ เขมิโย ป.ธ.๓) อดีต เจ้าคณะจังหวัดขอนแก่น (ธรรมยุต) วัดศรีจันทร์ อำเภอเมือง ได้นิมนต์พระอาจารย์สิงห์ ขนฺตฺยาคโม อาจารย์พระมหาปิ่น ปญฺญาพโล ป.ธ.๕ ซึ่งขณะนั้น อยู่ในระหว่างปฏิบัติศาสนกิจที่จังหวัดอุบลราชธานี มาช่วยศาสนกิจที่จังหวัดขอนแก่น พระอาจารย์สิงห์พร้อมด้วยพระอาจารย์กรรมฐานหลายรูป เช่น พระอาจารย์ฝั้น อาจาโร พระอาจารย์คูณ พระอาจารย์ดี พระอาจารย์อุ่น พระอาจารย์อ่อน พระอาจารย์สีโห พระอาจารย์สิม พระอาจารย์เพ็ง พระอาจารย์เกียรติ เป็นต้น มาตั้งวัด ณ บริเวณ โคกเหล่างา (ปัจจุบัน วัดวิเวกธรรม) ส่วนอาจารย์พระมหาปิ่น พร้อมด้วยพระอาจารย์เทศก์ เทสฺรํสี (พระราชนิโรธรังสี) วัดหินหมากเป้ง พระอาจารย์หลุย จนฺทสโร พระอาจารย์ภูมี พระอาจารย์สีลา พระอาจารย์กงมา และ พระอาจารย์แดง (พระสุธรรมคณาจารย์) วัดประชานิยม เป็นต้น ไปจำพรรษา ณ ดอนปู่ตา (ปัจจุบัน วัดสมศรี) บ้านพระคือ
กลางปีพุทธศักราช ๒๔๗๓ พระอาจารย์เทศก์ เทสฺรํสี พร้อมด้วยพระอาจารย์กรรมฐานหลายรูป ได้ออกมาพักปฏิบัติธรรม ณ บริเวณสถานที่สร้างวัดป่าแสงอรุณในปัจจุบัน ต่อมาอาจารย์พระมหาปิ่น ปญฺญาพโล ซึ่งมีอายุพรรษามากกว่าทุกรูปได้ออกมาจากดอนปู่ตา (วัดสมศรี) มาอยู่จำพรรษาร่วมกับพระอาจารย์เทศก์ จึงถือว่าผู้ที่เริ่มการบุกเบิกในการก่อตั้งและสร้างวัดป่าแสงอรุณครั้งแรก คือ พระอาจารย์เทศก์ และอาจารย์พระมหาปิ่น ดังที่ทราบอยู่โดยทั่วไปนั่นเอง
ครั้นปีถัดมา พ.ศ. ๒๔๗๔ อาจารย์พระมหาปิ่น ปญฺญาพโล ติดตามพระอาจารย์สิงห์ ขนฺตฺยาคโม และคณะพระอาจารย์กรรมฐาน ไปช่วยการพระศาสนาที่จังหวัดนครราชสีมา ตามคำนิมนต์ของท่านเจ้าคุณ พระธรรมปาโมกข์ จึงได้มอบหมายหน้าที่การบริหารวัดป่าพระคือ (ปัจจุบันคือวัดป่าแสงอรุณ) ให้พระอาจารย์ผู้มีพรรษารองลงมารับหน้าที่สืบต่อกันมาโดยลำดับ ปรากฏว่า วัดนี้มีพระอาจารย์กรรมฐานสายพระอาจารย์มั่น ภูริทตฺโต แวะพักและอยู่จำพรรษาให้การอบรม ในด้านการสมาธิภาวนา แก่พุทธบริษัทติดต่อกันมาไม่ขาดระยะ
- พ.ศ. ๒๕๑๘ เป็นที่ตั้ง กิจกรรมการส่งเสริม อนุรักษ์ ศิลปวัฒนธรรมท้องถิ่น เช่น การประกวดสรภัญญะ กลองยาว และแคน เป็นต้น จนถึงปัจจุบัน
- พ.ศ. ๒๕๒๖ เป็นสำนักศาสนศึกษาตัวอย่าง และเป็นสำนักศาสนศึกษาดีเด่นตามลำดับ
- พ.ศ. ๒๕๒๘ ได้รับพระราชทานวิสุงคามสีมา
- พ.ศ. ๒๕๓๐ เป็นที่ตั้ง สำนักงานเจ้าคณะจังหวัดขอนแก่น (ธรรมยุต)
