ทรงพระนามเดิมว่า ฉิม(สมเด็จพระเจ้าลูกยาเธอ เจ้าฟ้ากรมหลวงอิศรสุนทร) เป็นพระราชโอรสพระองค์ที่ 4 ของพระบาทสมเด็จพระพุทธยอดฟ้าจุฬาโลก ประสูติแต่สมเด็จพระอมรินทราบรมราชินี ณ บ้านอัมพวา แขวงเมืองสมุทรสงคราม เมื่อวันที่ 24 กุมภาพันธ์ พ.ศ.2310 เสวยราชสมบัติ เมื่อปีมะเส็ง ปี พ.ศ.2352 ขณะมีพระชนมายุได้ 42 พรรษา เป็นรัชกาลที่ 2 แห่งราชวงศ์จักรี และทรงครองราชย์จนถึงปี พ.ศ.2367 สิริดำรงราชสมบัติเป็นเวลา 15 ปี พระองค์ทรงมีพระราชโอรส พระราชธิดา รวมทั้งสิ้น 73 พระองค์

ปี พ.ศ.2349 พระองค์ทรงได้รับสถาปนาเลื่อนพระยศขึ้นเป็นกรมพระราชวังบวรสถานมงคล ต่อจากนั้นเป็นเวลาอีกเพียง 2 ปี พระบาทสมเด็จพระพุทธยอดฟ้าจุฬาโลกมหาราช พระบรมชนกนาถเสด็จสวรรคต พระองค์จึงได้เสด็จเถลิงถวัลย์ราชสมบัติเป็นพระมหากษัตริย์แห่งราชวงศ์จักรี ทรงพระนามว่า พระบาทสมเด็จพระพุทธเลิศหล้านภาลัย เมื่อวันที่ 7 กันยายน พ.ศ.2352

พระบาทสมเด็จพระพุทธเลิศหล้านภาลัย จึงทรงโปรดเกล้าฯ ให้สร้างเมืองและป้อมปราการต่างๆ ขึ้นเพื่อให้เป็นเมืองหน้าด่านคอยป้องกันข้าศึกที่จะยกเข้ามาทางทะเล ที่เมืองสมุทรปราการ และที่เมืองปากลัด (ปัจจุบันคือ อ.พระประแดง จ.สมุทรปราการ) โดยมีพระราชบัญชาให้กรมพระราชวังบวรมหาเสนานุรักษ์ เป็นแม่กองก่อสร้างเมืองนครเขื่อนขันธ์ขึ้นที่ปากลัด และสร้างปราการ 3 ป้อม คือ ป้อมปีศาจผีสิง ป้อมราหู และ ป้อมศัตรูพินาศ แล้วโปรดเกล้าฯ ให้อพยพครอบครัวชาวมอญจากปทุมธานี มาอยู่ที่นครเขื่อนขันธ์ด้วย
