กั้น ทองหล่อ เกิดเมื่อวันที่ 2 พฤศจิกายน พ.ศ. 2455 ที่บ้านเลขที่ 234 หมู่ที่ 2 บ้านน้ำกระจาย ตำบลพะวง อำเภอเมือง จังหวัดสงขลา เป็นบุตรคนที่ 2 ในจำนวน 5 คน ของนายคง และนางประชุม ทองหล่อ บิดาเป็นชาวบ้านน้ำกระจาย ส่วนมารดาเป็นชาวบ้านควนฝาละมี จังหวัดพัทลุง

กั้น ทองหล่อ อ่านเขียนตัวอักษรได้ก่อนเข้าเรียน พอทางการเปิดโรงเรียนประชาบาลขึ้นที่บ้านน้ำกระจาย เมื่อพ.ศ. 2467 กั้น ทองหล่อ ได้เข้าเรียนด้วย เรียนจบชั้นประถมปีที่ 4 เมื่ออายุ 16 ปี และเข้าเรียนต่อชั้น ม.1 ที่โรงเรียนมหาวชิราวุธ จังหวัดสงขลา แต่เรียนได้ปีเดียวก็ออกจากโรงเรียน เพราะสนใจและมีความต้องการที่จะไปประกอบอาชีพเล่นหนังตะลุง

ต่อมา กั้น ทองหล่อ เกิดท้อถอยในการแสดงหนังตะลุง และอยากเข้ารับราชการ จึงบรรพชาเป็นสามเณรที่วัดน้ำกระจาย เรียนทางธรรมอยู่สองปี ก็สอบนักธรรมโทได้ จึงลาสิกขามาเป็นครู บรรจุที่โรงเรียนน้ำกระจาย เป็นครูอยู่ได้ไม่ถึงปีก็ลาออกจากครูไปแสดงหนังตะลุงอีก เพราะชาวบ้านเห็นว่าหนังตะลุงดี ๆ แถบนั้นหายากมาก ผู้ที่เป็นตัวตั้งตัวตีอย่างแข็งขันให้ครูกั้นลาออกคือ หนังเอียด ปากพน ซึ่งมีศักดิ์เป็นอา อยู่บ้านเขาตก อำเภอกำแพงเพชร (ปัจจุบันคือ อำเภอรัตภูมิ จังหวัดสงขลา)

กั้น ทองหล่อ แสดงหนังตะลุงครั้งแรกเมื่ออายุ 18 ปี ชาวบ้านเรียกหนังกั้นว่า "หนังเด็ก" พออายุ 22 ปี ได้อุปสมบทที่วัดน้ำกระจาย ขณะที่เป็นภิกษุอยู่นั้นสามารถเทศน์มหาชาติได้ทั้ง 13 กัณฑ์ บวชได้พรรษาเดียวก็ลาสิกขา และประกอบอาชีพเป็นนายหนังตะลุงตั้งแต่นั้นเป็นต้นมา

หนังกั้น ทองหล่อ มีภรรยาสามคน คือ นางฉิ้น มีบุตร 1 คน นางกิ้มเลี่ยน มีธิดา 1 คน และนางซุ้ยเหียง มีบุตร 2 คน ธิดา 1 คน หนังกั้นถึงแก่กรรมเมื่อวันที่ 14 พฤษภาคม พ.ศ. 2531 อายุได้ 76 ปี

นายกั้น ทองหล่อ รักษาศิลปะในการแสดงหนังตะลุงแบบโบราณมาตลอด ไม่ว่าจะเป็นเครื่องดนตรี รูปหนังตะลุง และแนวการแสดง เรื่องที่ใช้แสดงหนังกั้นประพันธ์ขึ้นเองทั้งสิ้น ที่มีชื่อเสียงโด่งดังมากที่สุด คือเรื่องยอดชายนายนาบุญ การว่ากลอนก็เป็นกลอนปฏิภาณ แต่ละเรื่องที่แสดงจึงไม่มีกลอนซ้ำ รวมจำนวนครั้งที่หนังกั้นแสดง ทั้งในประเทศไทยและประเทศมาเลเซีย ประมาณ 6,000 ครั้ง

กั้น ทองหล่อ เป็นผู้ริเริ่มในการปรับปรุงการเล่นดนตรีแบบเล่นย้อนทับ (เล่นดนตรีให้เข้ากับเพลงปี่ แล้วเชิด แล้วหยุดนิ่ง) และนำเครื่องขยายเสียงมาใช้ในการเล่นหนัง หนังกั้นสามารถพากย์เสียงตามลักษณะของรูปหนังได้ไม่ซ้ำกันเลยไม่น้อยกว่า 14 เสียง และเป็นนายหนังที่มีลูกศิษย์มากที่สุด ซึ่งจะเห็นได้จากเมื่อการจัดงานพิธีไหว้ครูแก่หนังกั้น ที่บ้านน้ำกระจาย เมื่อปี พ.ศ. 2503 มีลูกศิษย์มาร่วมพิธีเป็นจำนวนมากกว่า 100 คน ลูกศิษย์หนังกั้นมาเล่นหนังฉลองถึงสามวันสามคืน

เมื่อพ.ศ. 2527 หนังกั้น ทองหล่อ ได้รับเชิญไปเป็นวิทยากรหลักที่ให้ความรู้เรื่องหนังตะลุงและประสบการณ์การเล่นหนังตะลุง ในการสัมมนาหนังตะลุง 14 จังหวัดภาคใต้ ที่วิทยาลัยครูสงขลา (ตอนนี้เป็น มหาวิทยาลัยราชภัฏสงขลา) และเป็นประธานที่ปรึกษาชมรมหนังตะลุงจังหวัดสงขลา เมื่อพ.ศ. 2528
