วัดสุปัฏนารามวรวิหาร ตั้งอยู่ที่ถนนวัดสุปัฏน์ อ. เมือง จ.อุบลราชธานี ( ริมฝั่งแม่น้ำมูล ซึ่งไหลผ่านทางทิศใต้ของตัวเมืองอุบลราชธานี ) มีเนื้อที่ 21 ไร่ 38 ตารางวา เป็นวัดธรรมยุติวัดแรกของจังหวัดอุบลราชธานี มีสถานะเป็นวัดอารามหลวงชั้นตรี
วัดสุปัฏนาราม สร้างในรัชสมัยของ พระบาทสมเด็จพระจอมเกล้าเจ้าอยู่หัว รัชกาลที่ 4 โดยได้เริ่มสร้างวัดในปี พ.ศ.2393 และสร้างเสร็จ พ.ศ.2396 พระบาทสมเด็จพระจอมเกล้าเจ้าอยู่หัว โปรดเกล้าฯ พระราชทานนามว่า "วัดสุปัฏนาราม อันหมายถึง วัดที่มีสถานที่ตั้งเหมาะสมเป็น ท่าเรือที่ดี " ซึ่งสภาพของที่ตั้งวัดขณะนั้นเป็นที่เงียบสงัด เหมาะที่จะบำเพ็ญศาสนกิจ สะดวกในการออกบิณฑบาต ที่ไม่ไกลอยู่ติดกับฝั่งลำน้ำมูล และเมื่อสร้างวัดเสร็จแล้ว จงไปอาธนา ท่านพนฺธุโล (ดี) มาครองวัดสุปัฏนาราม และ พระบาทสมเด็จพระจอมเกล้าเจ้าอยู่หัวทรงพระราชทาน ทรัพย์ส่วนพระองค์ ดังนี้

1. พระราชทานทรัพย์ส่วนพระองค์ 10 ชั่ง ( 800 ) บาท
2. ตั้งนิตยภัตแก่เจ้าอาวาส เดือนละ 8 บาท
3. จัดให้มีเลกวัด ( คนทำงานประจำวัด 60 คน)

สิ่งสำคัญภายในวัดสุปัฎนารามวรวิหาร

ต่อมาเจ้าอาวาสรูปที่ 7 สมเด็จพระมหาวีรวงศ์ (ติสฺโส อ้วน) สมัยดำรงสมณศักดิ์ที่พระพระราชมุณี เป็นเจ้าคณะมณฑลอุบลราชธานี จึงได้นิมนต์พระมหาเถระ ข้าราชการ คหบดี หลายฝ่ายได้เข้าร่วมประชุมปรึกษาหารือกัน และเห็นควรรื้อถอนพระอุโบสถหลังเก่าออก ปลูกสร้างใหม่ทดแทน ให้เกิดความมั่นคงถาวร โดยมีเหตุผล 2 ประการ คือ

1.ให้พ้นทาง หากมีการตัดถนนในอนาคต
2. ให้พ้นตลิ่งฝั่งแม่น้ำมูล โดยให้หันพระอุโบสถลงสู่แม่น้ำ (ทิศใต้)

ซึ่งพระอุโบสถมีขนาดกว้าง 20 เมตร ยาว 34 เมตร สูง 22 เมตร ผู้อำนวยการสร้าง ได้แก่ สมเด็จพระมหาวีรวงศ์ (ติสฺโส อ้วน) สถาปนิกผู้ออกแบบ คือหลวงสถิตย์นิมานกาล (ชวน สุปิยพันธ์) ซึ่งเป็นช่างหลวงเป็นผู้ออกแบบ โดยเริ่มเตรียมวัสดุก่อสร้างตั้งแต่ปี พ.ศ.2460

เนื่องจากการก่อสร้าง จะต้องสิ้นเปลืองวัสดุอุปกรณ์มากมาย และสมัยนั้น ยังไม่มีผู้ที่ทำไว้จำหน่ายเหมือนปัจจุบัน จึงได้ตั้งโรงงานช่างขึ้น แล้วสอนแบบก่อสร้าง สอนวิชาปั้นและก่อเตาเผาอิฐ สอนวิธีสีข้าว โดยวัดเป็นผู้ควบคุมดูแล เจ้าหน้าที่บ้านเมืองคือสมุหเทศบาล ผู้ว่าราชการจังหวัด และข้าราชการให้การสนับสนุนจน ได้รับความสำเร็จในที่สุด และการตั้งโรงงานนี้ นับว่าเป็นประโยชน์แก่ประชาชน เพราะเป็นจุดเริ่มต้น ที่ให้ความรู้ในการประกอบอาชีพ และประชาชนได้ยึดถือปฏิบัติต่อกันมา การก่อสร้างเสร็จในปี พ.ศ.2479 สิ้นค่าใช้จ่ายประมาณ 70,000 บาท

เมื่อสร้างเสร็จแล้ว จุดประสงค์แรกนั้นก็คือ ต้องการให้เป็นที่ประดิษฐานของพระสัพพัญญูเจ้า ซึ่งเป็นพระประธาน ในพระอุโบสถวัดสุปัฏนาราม และเนื่องจากพระอุโบสถหลังนี้ ค่อนข้างกว้างใหญ่ จึงใช้ประโยชน์ในการประกอบพุทธศาสนพิธี ไม่ว่าจะเป็นพิธีกรรมของสงฆ์โดยตรง หรือพิธีกรรมที่มีพุทธศาสนิกชนร่วมด้วยก็ตาม
