ตั้งอยู่ภายในตำบลเกาะพระทอง มีพื้นที่ 80,000 ไร่ และได้ประกาศเป็นเขตอุทยานแห่งชาติ ในวันที่ 1 กันยายน พ.ศ. 2525 ส่วนชื่อของ "เกาะสิมิลัน" สิมิลันเป็นภาษายาวี หรือ มลายู แปลว่าเก้า หรือ หมู่เกาะเก้า เป็นหมู่เกาะเล็ก ๆ ในทะเลอันดามันมีทั้งหมด 9 เกาะ เรียงลำดับจากเหนือมาใต้ ได้แก่ เกาะหูยง, เกาะปายัง, เกาะปาหยัน, เกาะเมี่ยง (มี 2 เกาะติดกัน), เกาะปายู, เกาะหัวกะโหลก (เกาะบอน), เกาะสิมิลัน และเกาะบางู เป็นต้น ส่วนบริเวณที่ทำการอุทยานฯ อยู่ที่เกาะเมี่ยง เพราะเป็นเกาะที่มีน้ำจืด ส่วนเดือนพฤศจิกายนถึงเดือนเมษายนเป็นช่วงที่น่าท่องเที่ยวมากที่สุด และเดือนพฤษภาคมถึงพฤศจิกายนเป็นฤดูมรสุมตะวันตกเฉียงใต้ มีคลื่นลมแรง เป็นอันตรายต่อการเดินเรือ ทางอุทยานฯ จะมีการประกาศปิดเกาะในเดือนพฤษภาคมเพื่อเป็นการฟื้นฟูธรรมชาติทุกปี
