พื้นที่เขตรักษาพันธุ์สัตว์ป่าดงใหญ่ อยู่ในพื้นที่ป่าสงวนแห่งชาติป่าดงใหญ่ ท้องที่ตำบลนางรอง ตำบลโนนดินแดง กิ่งอำเภอโนนดินแดง ตำบลโคกมะม่วง ตำบลหูทำนบ อำเภอปะคำ จังหวัดบุรีรัมย์ มีเนื้อที่ 212,500 ไร่ หรือประมาณ 340 ตารางกิโลเมตร เพื่อให้เป็นที่อยู่อาศัย หลบภัย ขยายพันธุ์ และประกอบกิจกรรมของสัตว์ป่าโดยปลอดภัย ช่วยรักษาพันธุ์ไม้ชนิดต่าง ๆ ไว้ และยังช่วยรักษาแหล่งต้นน้ำลำธาร ซึ่งไหลหล่อเลี้ยง ชีวิตประชาชนชาวบุรีรัมย์ให้คงอยู่ตลอดไป


เมื่อวันที่ 14 กรกฎาคม พ.ศ. 2548 อุทยานแห่งชาติเขาใหญ่ ทับลาน ปางสีดา ตาพระยา และเขตรักษาพันธุ์สัตว์ป่าดงใหญ่ ได้รับการประกาศให้เป็น "มรดกโลกทางธรรมชาติ" จากองค์การยูเนสโก ภายใต้ชื่อกลุ่ม “ดงพญาเย็น-เขาใหญ่”
