โรงเรียนปราจิณราษฎรอำรุง เดิมเป็นโรงเรียนชายล้วน เปิดทำการสอนเมื่อวันที่ 1 พฤศจิกายน พ.ศ. 2456 ในระดับชั้นเรียนประถมศึกษาเป็นลำดับแรก ต่อมามีจำนวนนักเรียนเพิ่มมากขึ้น จึงได้สร้างอาคารเรียนถาวรไม้ชั้นเดียวใต้ถุนสูงขึ้น เมื่อ พ.ศ. 2460 และเปิดเรียนถึงชั้นมัธยมปีที่ 6 ได้เริ่มเป็นสหศึกษาโดยการรับ มัธยมตอนปลายก่อน แล้วจึงเริ่มเต็มรูปแบบ

ต่อมาในปี พ.ศ. 2478 กระทรวงกลาโหมได้ยกที่ดินให้กระทรวงศึกษาธิการ และได้ก่อสร้างอาคารเรียนมีใต้ถุนสูงขนาด 6 ห้องเรียน เพิ่มเติม และย้ายนักเรียนมัธยมศึกษามาเรียนที่อาคารหลังใหม่นี้ ส่วนโรงเรียนที่ตั้งอยู่บริเวณเดิมได้ใช้เป็นที่เรียนชั้นประถมศึกษาต่อไป ซึ่งต่อมาภายหลังได้ตั้งเป็นโรงเรียนปราจีนกัลยาณีซึ่งเป็นโรงเรียนหญิงล้วน

ปัจจุบันเป็นโรงเรียนประจำจังหวัดปราจีนบุรีเป็นที่รู้จักในนาม ป.ร.อ.
