วัดพระศรีสรรเพชญตั้งอยู่ในเขตพระราชวังโบราณเป็นวัดพุทธาวาสที่ไม่มีพระสงฆ์จำพรรษาเพื่อประกอบพิธีสำคัญต่างๆของเมือง

วัดพระศรีสรรเพชญ์ เดิมในสมัยสมเด็จพระรามาธิบดีที่ 1 ใช้เป็นที่ประทับ ต่อมาสมเด็จพระบรมไตรโลกนาถ ทรงสร้างพระราชมณเฑียรขึ้นใหม่ทางตอนเหนือ แล้วจึงโปรดฯให้ยกเป็นเขตพุทธาวาส เพื่อประกอบพิธีสำคัญต่าง ๆ ของบ้านเมือง จึงเป็นวัดในเขตพระราชวังที่ไม่มีพระสงฆ์จำพรรษา แตกต่างกับวัดมหาธาตุสุโขทัย ที่มีพระสงฆ์จำพรรษา ทั้งวัดมหาธาตุ สุโขทัย,วัดพระศรีสรรเพชญ์ อยุธยา และวัดพระศรีรัตนศาสดาราม ต่างก็ถูกสถาปนาขึ้นในมูลเหตุการสร้างวัดเดียวกันนั่นคือ "สร้างเพื่อเป็นวัดประจำพระราชวัง"
