พิพิธภัณฑสถานแห่งชาติ เจ้าสามพระยา สร้างขึ้นเนื่องในวโรกาสที่พระบาทสมเด็จพระเจ้าอยู่หัว และสมเด็จพระนางเจ้าพระบรมราชินีนาถ เสด็จพระราชดำเนินมาทอดพระเนตรโบราณวัตถุที่พบจากกรุพระปรางค์วัดราชบูรณะ จังหวัดพระนครศรีอยุธยา เมื่อวันที่ 23พฤศจิกายน พ.ศ. 2500 ทรงมีพระราชปรารภกับรัฐมนตรีกระทรวงศึกษาธิการและอธิบดีกรมศิลปากรในสมัยนั้นว่า “ โบราณวัตถุ และศิลปวัตถุที่พบในกรุพระปรางค์วัดราชบูรณะนี้ สมควรจะได้มีพิพิธภัณฑสถานเก็บรักษา และตั้งแสดงให้ประชาชนในจังหวัดพระนครศรีอยุธยานี้ หาควรนำไปเก็บรักษา และตั้งแสดง ณ ที่อื่นไม่” กรมศิลปากรจึงได้สร้างพิพิธภัณฑสถานแห่งชาติ เจ้าสามพระยา ขึ้นเพื่อเก็บรักษาจัดแสดงโบราณวัตถุที่พบจากกรุพระปรางค์วัดราชบูรณะ และโบราณวัตถุพบในจังหวัดพระนครศรีอยุธยา อาคารก่อสร้างด้วยเงินบริจาคจากประชาชน ซึ่งผู้บริจาคได้รับพระพิมพ์ที่พบจากกรุพระปรางค์วัดราชบูรณะเป็นการสมนาคุณ ทรงพระกรุณาโปรดเกล้าฯ ให้อัญเชิญพระนามสมเด็จพระบรมราชาธิราชที่ 2 (เจ้าสามพระยา) ผู้ทรงสร้างพระปรางค์วัดราชบูรณะเป็นนามพิพิธภัณฑ์
